正常及脑缺血-再灌注大鼠静脉注射清开灵后黄芩苷在脑、肝、肺中的分布

摘    要:目的研究清开灵注射液(QKL)中黄芩苷(BC)在生理及病理大鼠效应器官(脑、肝、肺)分布的不同。方法取正常和脑缺血-再灌注大鼠,尾静脉注射QKL 800 mg.kg-1后,于不同时间点处死,取脑、肝、肺组织,HPLC-MS测定BC含量。结果静脉给药后,正常大鼠肺中BC含量明显高于肝、脑,脑部浓度最低;脑缺血-再灌注大鼠脑中BC浓度较正常大鼠明显升高,肝中分布也有显著增加,但肺中BC浓度较正常组明显降低。结论QKL中BC在正常及病理大鼠的效应器官均有分布,但两种状态下分布具有明显区别,脑缺血-再灌注状态有利于BC快速透过血-脑屏障。

Abstract:OBJECTIVE To determine the active distribution of baicalin in Qingkailing injection in rats' brain,lung and liver under physiological and pathological condition.METHODS Qingkailing was injected by caudal vein to both normal and cerebral ischemia reperfusion rats.The brain,liver and lung were collected at various time,and the concentration of baicalin from samples was determined by HPLC-MS.RESULTS After iv Qingkailing injection,the concentration of baicalin in the lung was far higher than in the liver and brain,the concentration of baicalin in the brain was minimum in normal rats.In cerebral ischemia reperfusion rats,the concentration of baicalin in the brain was higher than that of normal rats,the distribution in the liver increased,but the distribution in the lung decreased.CONCLUSIONS In both normal and cerebral ischemia reperfusion rats,baicalin was distributed to the brain,the liver and the lung.There was obvious difference under different conditions,cerebral ischemia reperfusion was in favor of baicalin's quickly permeation through the blood-cerebral barrier.
