摘 要: | 目的研究刺五加皂甙(ASS)对青霉素致痫后大鼠海马长时程增强(LTP)的影响。方法实验分为侧脑室注射(icv)和腹腔注射(ip)两部分,每部分有NS对照组、模型对照组(青霉素致痫组)、ASS小、中、大剂量组。观察不同组别海马CA1区群峰电位(PS)幅度的差异,以此探讨ASS对癫痫大鼠海马LTP的作用。结果模型对照组海马PS幅度显著小于NS对照组(P〈0.01或P〈0.05);ASS侧脑室及腹腔注射组PS幅度显著大于模型对照组(P〈0.05或P〈0.01);ASS(icv)中剂量组PS幅度比小、大剂量组大(P〈0.01),小、大剂量组之间无差异;ASS(ip)小、大剂量组之间无差异。结论ASS(icv)对青霉素致痫大鼠海马LTP抑制有明显改善作用,但大剂量ASS改善作用减弱;ASS(ip)对大鼠海马LTP抑制有改善作用,但可能由于血脑屏障的存在,无量效关系。

Abstract: | Objective The purpose of our works was to study effects of Acantbepanax Senticsus Saponin (ASS) on the LTP of hippocampus in epileptic rats induced by penicillin. Methods The experiment consists of intracerebroventricular injection and intraperitoneal injection parts ,each part includes NS group,epilepsy model group,low does of ASS group,medium does of ASS group and high does of ASS group. The effects were evaluated by the amplitude of population spike(PS) in Hippocampal CA1 section of these groups. Results Amplitude of PS in epilepsy group is apparently lower than that in NS group,amplitude of PS in ASS(icv&ip) groups is apparently bigher than that in that in epilepsy group(P 〈 0.01 ) ;and that medium does group of ASS(icy) higher than low and high does groups(P 〈 0. 01 ), low and high does group have no difference; no significant difference exists between low and high does of ASS(ip) groups. Conclusion ASS (icv) could reverse LTP depression in epileptic rats induced by penicillin, but high does makes less effects;ASS(ip) reverse LTP depression in epileptic rats induced by penicillin, but hemato encephalic barrier may be the cause of nondifference between low and high does. |
