摘    要:目的:建立别嘌醇高效液相色谱法有关物质测定。方法:采用HPLC—Uv法,并考察色谱条件,优化分离效果,以十八烷基硅烷键合硅胶为填充剂;以甲醇-1.25班磷酸二氢钾溶液(20:80)为流动相;检测波长为230nm。结果:杂质能与别嘌醇得到有效分离,分离度大于1.5;检测限为0.6ng,精密度良好(RSD为1.89%)。结论:高效液相色谱法可用于测定别嘌醇中的有关物质。

Abstract:Objective:To establish a reversed-phase high performance iiquid chromatography (RP-HPLC) for determination of allopurinol related substances. Methods :The specific and stable RP-HPLC with ultraviolet detection was employed,and the chromatographic conditions were investigated to optimize the separation. The chromatography was accomplished on C18, and the moble phase con- sisted of methanol-1.25 g/L potassium dihydrogen phosphoric acid,and the detection wavelength was set at 230 nm. Results:All the related substances gained a completely chromatographic separation with the principal peak.The limit of detection(LOD) was 0.6 ng based on S/N greater than 3:1.The sensitivity was much good(RSD〈1.89%. Conclusion:The method is adopted for determination of allopurinol related substances.
