川芎嗪干预大鼠实验性肝纤维化的研究

目的研究川芎嗪对大鼠实验性肝纤维化的治疗作用,并探讨其剂量-效应关系。方法90只SD雄性大鼠随机取80只以40%四氯化碳皮下注射,其余10只予以相同容量生理盐水皮下注射作为正常对照组(N组)。8周后,造模组随机处死16只,经肝组织活检证实肝纤维化形成(模型对照组,M组)。剩余肝纤维化大鼠随机分成3组:分别为川芎嗪低剂量治疗组(T1组,川芎嗪一日1次20mg/kg腹腔内注射)、高剂量治疗组(T2组,川芎嗪一日1次80mg/kg腹腔内注射)、空白对照组(R组,生理盐水一日1次腹腔内注射),疗程8周。试验结束时,全部大鼠采血后处死,分别行HE染色评估肝纤维化程度、Masson染色评估肝组织中胶原纤维百分比、生化法检测血清谷丙转氨酶及谷草转氨酶水平。结果T1、T2组大鼠肝纤维化程度较R组明显减轻,胶原面积密度降低,血清转氨酶下降,且T2组改善优于T1组。结论川芎嗪具有逆转肝纤维化的作用,且高剂量疗效更显著。

Experimental Studies on the Therapeutic Effects of Ligustrazine on Liver Fibrosis in Rats

Objective To investigate the therapeutic effects of ligustrazine on experimental liver fibrosis in rats,and to explore the relationship between the dosage and the effects.Methods After one week of normally feeding,liver fibrosis was induced in 80 SD rats by using subcutaneous injection of 40% CCI_4,and the other 10 rats were routinely fed as normal group (Group N).After 8 weeks,the remaining rats were divided randomly into 4 groups;model group (Group M),treatment group with a lower dose(Group T1),treatment group with a higher dose(Group T2),and recovery group(Group R).The rats of 2 treatment groups were given ligustrazine for 8 weeks.The stage of liver fibrosis was assessed through HE staining,and the density of collagen in liver speciments was determined through Masson staining.Results Compared with R group,the stage of hepatic fibrosis in T1 and T2 group was obviously reduced and the area density of collagen in liver tissues in T1 and T2 group was obviously lower(P<0.05).The higher dosage seems more effective in treatment of hepatic fibrosis in rats. Conclusions Ligustrazine has obvious therapeutic effects on rats with liver fibrosis.The higher dosage of ligustrazine is more effective.
