| 摘 要： | 目的:观察镰形棘豆对人肝癌细胞SMMC-7721的增殖及其基质金属蛋白酶-2(MMP-2)分泌量的影响.方法:以不同质量浓度(25,50,100,250,500 mg·L-1)的镰形棘豆挥发油、总多糖、总黄酮、总生物碱和总皂苷部位处理SMMC-7721细胞24 h后,采用MTT法检测各部位对SMMC-7721细胞增殖的抑制作用;采用ELISA法测定挥发油部位和总黄酮部位处理SMMC-7721肝癌细胞24 h后细胞上清中MMP-2的分泌量;Real-time PCR法检测处理24 h后MMP-2 mRNA的转录情况.结果:MTT实验结果表明,挥发油和总黄酮部位抑制SMMC-7721细胞的增殖,并表现出一定的浓度依赖性.ELISA实验表明挥发油部位显著性地抑制MMP-2分泌量;Real-time PCR实验表明挥发油和总黄酮均显著抑制MMP-2 mRNA的表达.结论:镰形棘豆挥发油和总黄酮部位抑制人肝癌细胞SMMC-7721的增殖活性,可能与其下调MMP-2的分泌量和转录水平有关.
