摘    要:目的:建立同时测定五味子浸膏中五味予醇甲、五味子甲素与五味子乙素的含量的反向高效液相色谱(RP-HPLC)法。方法:色谱柱为Diamonsil(R)钻石C18柱(200×4.6mm,5um),流动相为甲醇-水(梯度洗脱)。紫外检测波长250nm,柱温30℃,流速1.0ml/min。结果:五味子醇甲、五味子甲素、五味子乙素分别在0.732—5.612ug(r=0.9999)、0.171—1.311ug(r=0.9999)、0.354~2.714ug(r=0.9999)的范围内与峰面积积分值线性关系良好。平均回收率五味子醇甲为99.86%,RSD值为1.76%;五味子甲素为99.53%,RSD值为0.85%;五味子乙素为99.61%。RSD值为0.96%。结论:本法准确,快速,灵敏度高,可用于五味子浸膏中有效成分的定量分析。

Determination of Three Composition in Compound Chinese Magnoliavine Fruit Extract by HPLC

Abstract:Objective: To establish a method for content determination of Schisandrin, Deoxyschizandrin and Schisandrin Bin Chinese magnoliavine fruit extract by RP - HPLC. Methods:The Diamonsil C18 column(200 mm ×4. 6 mm,5 um) was used, mobile phase of water and McOH (gradient elution)was set up, the flow rate was 1.0 ml/min, the column temperature was 30℃ and the detection wavelength was 250nm. Results:The linear ranges of Schisandrin,Deoxyschizandfin and Schisandrin B were within 0. 732 - 5.612ug ( r = 0. 9999) ,0. 171 - 1.311 ug ( r = 0. 9999) and 0. 354 - 2. 714ug ( r = 0. 9999), respectively. The average recoveries were 99.86% ( RSD 1.76% ) ,99. 53% ( RSD 0. 85% ) and 99. 61% ( RSD 0. 96% ), respectively. Conclusion:The established method is accurate, reliable and highly specific, and it can be used for the quality contrel of compound Chinese magnoliavine fruit extract.
