微量板倍量稀释法检测IgG抗A(B)效价改进研究

摘    要:目的通过实验研究改进微量板倍量稀释法检测IgG抗A(B)效价。方法采用微量板倍量稀释法,选取一个高值和一个低值标本来比较裂解液类型(2-Me、DTT)与稀释比例、裂解液处理时间和细胞温浴时间差异对结果的影响,综合得出最佳操作方案,将此方案与试管倍量稀释法进行比较,判断改进后的效果。结果得出改进后的操作方案:DTT与血清比例为3∶1,37℃处理时间为60min,细胞温浴时间为30min。与试管倍量稀释法比较无显著性差别(P>0.05)。结论改进后的方法是准确、有效的,更适合实验室进行大批量标本检测。

Abstract:Objective To modify microplate serial twofold dilution test in determining the titer of anti-IgG A/B antibody.Methods We chose one high value and one low value samples to mensurate the titers in different lysis solution(2-Me、DTT),dilution ratios,lysis times,and inbubating times by microplate serial twofold dilution test.And we obtained an optimal modified method.Then the modified method was compared with tube serial twofold dilution method.Results The optimal modified method was the follewing: the ratio of DTT to serum was 3 to 1;samples were treated at 37℃ in 60 minutes;cells incubated in 30 minutes.There was no significant difference(P>0.05) between two methods.Conclusion The modified method was accurate and effective,and it was more suitable for detecting a large number of samples on laboratory.
