芪棱汤对大鼠脑缺血再灌注后nestin蛋白表达的影响

目的观察芪棱汤对脑缺血再灌注后nestin蛋白表达的影响,为益气养阴活血法治疗缺血性中风提供实验依据。方法将大鼠随机分为假手术组、模型组、芪棱汤去养阴药组及芪棱汤组,采用颈内动脉线栓加环扎法复制大鼠脑缺血再灌注模型。于再灌注后不同时间点进行神经学评分,用免疫组化法检测nestin蛋白表达。结果假手术组神经学评分均正常,其余各组大鼠神经功能评分差异均有显著性,芪棱汤组明显优于其他组;假手术组未见nestin蛋白表达,模型组、芪棱汤去养阴药组和芪棱汤组nestin蛋白表达差异有显著性,芪棱汤组nestin蛋白表达明显高于其他各组。结论芪棱汤可以促进nestin蛋白表达,从而对缺血再灌注所导致的脑损伤起保护作用。

Influence of Qileng Decoction on Expression of Nestin Protein after Focal Cerebral Ischemic Reperfusion in Rats

Objective:To explore the influence of Qileng decoction on expression of Nestin protein after focal cerebral ischemic reperfusion in rats, and to provide the scientific foundation for benefit vital energy(BVE), nourishing yin(NY),activating blood circulation(ABC)on treating ischemic stroke.Methods:The rats were randomly divided into four groups including sham-operation group, model group, Qilengquyangyingyao(QLQYYY)decoction group and Qileng decoction group. The model of focal cerebral ischemia reperfusion injury were was made by using thread embolizing and fastening of the ICA in rats. Neurography grade were evaluated. Nestin protein were observed by immunohistochemical method at different time point after cerebral ischemia reperfusion.Results:The neurography grade of sham-operation group were normal. There was statistic significance among model group, QLQYYY decoction group and Qileng decoction group in evaluation of neurography grade. The effect of Qileng decoction group was superior to that of other groups. The expression of Nestin protein was not found in sham-operation group. There was statistic significance among model group, QLQYYY decoction group and Qileng decoction group in expression of Nestin protein,and The effect of Qileng decoction group was superior to that of other groups. Conclusion:Qileng decoction plays the neuron protective effects after cerebral ischemic reperfusion by promoting the expression of the Nestin protein. It may be one of the mechanism of BVE-NY-ABC treating ischemic stroke.
