急性坏死性胰腺炎大鼠硫氧还蛋白-1肺表达的实验研究

摘    要:目的:探讨内源性硫氧还蛋白一1(Thioredoxin-1,Trx-1)在急性坏死性胰腺炎(AcuteNecrotizingPancreatitis,ANP)大鼠模型肺组织中的表达。方法:将48只SD大鼠随机分成对照组(C组,n=24)、急性胰腺炎组(A组,n=24)。A组用6%的左旋精氨酸(L-Arginine,L—Arg)腹腔内注射,每次1.5mg/g体重,共3次,注射间隔1h,诱导ANP模型;C组同法注射等量生理盐水。在注射完L—Arg后的第6小时、第12小时和第24小时3个时点分批处死大鼠,应用免疫组织化学技术检测各组ANP肺组织Trx-1的表达,并观察各组对应各时点胰腺和肺组织病理学改变的变化。结果:①A组大鼠胰腺和肺组织病理损伤在各时点比C组明显加重(P〈O.01);②A组肺组织Trx-1各时点表达较C组明显增强(Pd0.01);③在第24小时,A组血清淀粉酶较C组显著升高(Pd0.01);④A组胰腺病理损伤与血清淀粉酶、肺损伤评分、肺免疫组化评分呈正相关(rx=0.374,P=0.046;rx=0.452,P=0.027;n=0.500,P=0.013)。结论:在ANP肺组织损伤程度与胰腺组织损伤程度密切相关。在ANP肺损伤组织中Trx-1表达显著增加,肺组织Trx-1过度表达与急性胰腺炎相关性肺损伤关系密切。

EXPRESSION OF TRX-1 IN RAT LUNGS WITH ACUTE NECROTIZING PANCREATITIS

Abstract:Objective:To study the expression of thioredoxin-l(Trx-1) in rat lungs with L-arginine(L-Arg)- induced acute necrotizing pancreatitis(ANP). Methods:Forty-eight Spraque-Dawley male rats were randomly divided into groups A and C. ANP was induced by injected intraperitoneally three times with 1. 5 mg/g body weight 6~ L-Arg in an interval of 1 hour in group A. Group C received saline alone with the samemethods. Rats were killed at the 6th,12th and 24th hours after the last time of L-Arg injection. The expression of Trx-1 in the lung tissues was detected with the method of immunohistochemistry. The patho- logical changes of pancreatic and lung tissues were respectively analyzed and scored according to Kusser's and Lei weizhang's standard. Results:OWith time going,the pathological injuries of the pancreas and lungs in group A were more serious than those in group C ( P 〈0.01). (1)The expression of Trx-1 in lungs in group C was significantly lower than that in group A ( P 〈0.01). (3)At the 24th hour,the serum amylase in group A was significantly higher than that in group C ( P 〈0.01). (4)Pathological injury score in pancreas was correlated with serum amylase, the lung injury and the lung immunohistochemistry score in Group A ( rs =0. 374, P =0. 0463 rs =0. 452, P =0. 027; rs =0. 500, P =0. 013). Conclusion:Lung injury is closely related to pancreatic tissue injury in ANP. The expression of Trx-1 in the lung injury increases significantly in ANP. Overexpression of Trx-1 in lung tissues associates closely with acute pancreatitis-associated lung injury.
