摘    要:传统的考马斯亮蓝R250(CBBR250)染色方法存在背景深,耗时长,试剂耗用多的缺点,考马斯亮蓝G250(CBBG250)虽然需时短,但其灵敏度效低。本文探讨了一步性低背景CBBR250染色方法在蛋白质PAGE中的应用,结果表明使用乙醇-乙酸-水染色系统,CBBR250的最佳工作浓度为0.004‰~0.008‰,染色时间为8~10h,灵敏度为0.5μg/带。一步性CBBR250染色方法与传统的CBBR250染色方法相比,有需时短,背景浅,试剂用量少的优点,可用于蛋白质的定性和定量研究。

Abstract:This paper studied a new method for protein staining with CBBR_250 in PAGE which had shallower background,and needed less time and reagents than the traditional method.The result showed that in the staining system with alcohol-acetic acid-H20 as solvent,the optimal working concentration of CBBR250 was from 0.004‰ to 0.008‰,the recommended staining time was 8~10 hours,and the sensitivity was 0.5μg protein per electrophoresis zone.
