葛根素预处理对大鼠脑缺血再灌注损伤后脑组织钙含量的影响

摘    要:目的观察葛根素预处理对大鼠局灶性脑缺血再灌注损伤后脑组织钙含量的影响,并探讨其可能的发生机制,为葛根素的临床应用提供依据。方法制作大鼠大脑中动脉缺血2 h后再灌注损伤模型。SD大鼠72只,随机分为假手术组?缺血再灌注组和葛根素预处理组,依术后处死动物时间的不同,每组再分为3个亚组,每个亚组8只动物。采用全自动生化分析仪测定脑组织钙含量。结果缺血再灌注组术后24 h、72 h、120 h各时间点钙含量均高于假手术组(P<0.05);葛根素预处理组24 h、72 h、120 h各时间点钙含量均低于同期缺血再灌注组(P<0.05)。结论葛根素预处理能显著减少大鼠局灶性脑缺血后脑组织钙含量,对脑组织起保护作用。

Abstract:Objective To observe the influence of puerarin preconditioning on calcium content in brain tissues of rats with cerebral ischemical reperfusion injury,to probe its potential genetic mechanism,and to provide evidences for puerarin clinical application.Methods Ischemic middle cerebral artery occlusion(MCAO) was taken 2 h before the rat models were treated with reperfusion injury.Seventy-two Spranue-Dawley(SD) rats were randomly divided into sham operation group(SO group),ischemia reperfusion group(IR group) and puerarin preconditioning group.Each group was divided into three subgroups according to the time of killing the rats,8 rats in each subgroup.Calcium content was determined by automatic biochemistry analyzer.Results Twenty four hours,72 h,and 120 h after the surgery,Calcium contents of the IR group were higher than those of the SO group,respectively(P<0.05);and also higher than those of the puerarin preconditioning group(P<0.05).Conclusion Puerarin preconditioning can significantly reduce calcium content in murine brain with focal cerebral ischemia and can protect brain tissues.
