TRAP-银染色法用于肝癌组织端粒酶活性测定的研究

目的探讨端粒酶活性检测在肝癌诊断中的意义。方法用裂解液提取组织细胞中的端粒酶模板,在特异的引物作用下进行PCR扩增,所得产物作聚丙稀酰胺凝胶垂直板电泳,经硝酸银染色分析端粒酶活性。结果肝癌组织中的端粒酶阳性率为85.7%,明显高于慢性肝炎组2.6%、肝硬化组16.7%和对照组4.2%,P<0.01。慢性肝炎组、肝硬化组、对照组之间无显著性差异(P<0.05)。结论端粒酶活性检测对临床恶性肝脏肿瘤疾病的诊断具有良好的应用前景。

Detection of Liver Cancer Telomerase Activity Using Telomeric Repeat Amplification Protocol Silver Stain

Objective To investigate the significance of telomerase activity detection in liver tumor diagnosis. Methods Telomerase template was extracted from the liver tissues and cells by lysis buffer, PCR amplification was performed under specific primer elongation. The PCR products were electrophoresed by vertical plate on 12% invariance polyacrylamide gel. Telomerase activity was analyzed using sliver stain. Result The positive rate of telomerase was 85.7% of patients with liver cancer tissues higher than 2.6% of patients with chronic hepatitis and 16.7% of hepatocirrhosis patients and 4 2% of control tissues obviously(P<0.01). The patients results of chronic hepatitis and hepatocirrhosis and control tissues did not differ obviously(P>0.05) Conclusion TRAP-silver stain protocol is sensitive and repetitive well. Testing telomerase is applicable in the diagnosis of clinical liver cancer.
