骨关节炎关节液促进滑膜间充质干细胞软骨分化

目的 观察骨关节炎(OA)关节液对滑膜间充质干细胞(SDSCs)软骨分化能力的影响.方法 用OA患者手术切除的滑膜组织标本分离培养SDSCs并诱导成软骨分化.将OA患者的关节液加入诱导后的SDSCs培养液中,模拟体内的OA关节环境继续培养(实验组),以不加OA关节液的SDSCs作为对照组.测量两组SDSCs形成的软骨小球的直径,采用实时荧光定量PCR和蛋白质印迹法检测成软骨相关标志物COLⅡ、Aggrecan、SOX9的mRNA及蛋白表达.结果 OA关节液处理16 d后,实验组软骨小球直径大干对照组(P＜0.05),COLⅡ、Aggrecan、SOX9 mRNA和蛋白的表达均高于对照组(P＜0.05).结论 OA关节液能够促进SDSCs软骨分化.

Synovial fluid of osteoarthritis promotes the differentiation of synovial mesenchymal stem cells

Objective To investigate the effect of OA on the differentiation of SDSCs. Methods in vitro experiments, the OA joint fluid was added to the SDSCs culture medium, and the effect of OA joint fluid on the differentiation ability of SDSCs was observed. Results The observation of the naked eye showed that the diameter of the cartilage in the experimental group was greater than that in the control. And it was observed that the expression of II COL, Aggrecan and SOX9 gene in the experimental group was significantly higher than that in the control group, and the II SOX9, Aggrecan and COL in the experimental group were significantly higher than those in the control group. Conclusion The results of this study confirm that the OA joint fluid in the whole has a role in promoting the differentiation of SDSCs.
