银丹心脑通软胶囊对VD大鼠学习记忆能力及AchE、NR2B的影响

摘    要:目的:研究银丹心脑通软胶囊对血管性痴呆模型大鼠学习记忆能力及海马CA1区乙酰胆碱酯酶(AchE)、N-甲基D-天(门)冬氨酸受体2B(NR2B)的影响。方法:采用双侧颈总动脉结扎,缺血、再灌注,同时腹腔注射硝普钠方法制备模型,采用Morris水迷宫实验对大鼠的学习记忆能力进行测试,用HE染色观察病理变化,免疫组化染色技术对大鼠海马CA1区AchE及NR2B进行检测。结果:假手术组与正常对照组比较学习记忆能力无差异,海马CA1区AchE及NR2B表达较模型组、治疗组少;模型组与正常对照组比较学习记忆能力明显下降,海马CA1区AchE及NR2B表达增多,差异有统计学意义(P〈0.05);治疗各组与模型组比较银丹心脑通软胶囊中剂量组和高剂量组学习记忆能力明显改善,海马CA1区AchE及NR2B表达减少,差异有统计学意义(P〈0.05)。结论:银丹心脑通软胶囊可改善VD模型大鼠的学习记忆能力,机制可能是由于降低海马CA区AchE及NR2B的表达。

Effect of Yindan Xinnaotong Capsule on learning and memory ability and AchE/NR2B in the vascular dementia rat

Abstract:Objective:To study the effect of Yindan Xinnaotong Capsule on learning and memory ability and AchE and NR2B in the hippocampus CA1 area of rats with vascular dementia.Methods:The rats model was established by bilateral common carotid artery blocking,repeated cerebral ischemia and reperfusion in combination with intraperitoneal injection of sodium nitroprusside.Learning and memory abilities of rats were assessed by Morris water maze(MWM) task,pathological changes were examined by HE staining,and AchE and NR2B in the hippocampal CA1 region were tested by immunohistochemical assay.Results:No significant difference was found in rats from normal group and sham operation group concerning rat's memory ability while AchE and NR2B of the rats in the hippocampal CA1 region in normal group and sham operation group were fewer than that of the rats in model control group and treatment groups;Compared with the rats in normal group,the memory ability of rats in model control group declined,while AchE and NR2B in the hippocampus CA1 region of rats increased(P0.05);Compared with that of the rats in model group.Rats' memory ability in treatment group with middle and large dose of Yindan Xinnaotong Capsule had markedly improved while AChE and NR2B in the hippocampus CA1 region of rats decreased(P0.05).Conclusion:Yindan Xinnaotong Capsule can significantly improve the memory of rat which may be related to its effect of reducing AChE in hippocampus.
