黄芪、尼莫地平对大鼠脑缺血再灌注损伤后神经细胞凋亡的影响

摘    要:目的:研究黄芪、尼莫地平单独及联合应用对大鼠脑缺血再灌注时神经细胞凋亡的影响。方法:建立SD大鼠脑缺血再灌注模型,将60只SD大鼠随机分为5组,未手术组(0组)予以正常剂量麻醉剂;对照组(1组)予以0.9%NaCl溶液2ml;黄芪治疗组(2组)予以黄芪注射液6ml/kg,分别于术前12h及30min,术后每隔12h注射1次,直至48h后;尼莫地平治疗组(3组),缺血前30min给予尼莫地平;黄芪联合尼莫地平治疗组(4组),缺血前予以尼莫地平、手术后予黄芪,每组各12只。每组使用不同药物后进行对照,采用原位末端凋亡荧光法检测凋亡细胞。结果:0、1、2、3、4组凋亡细胞数分别为0.8±0.4、48.5±11.3、33.5±10.2、30.2±8.6、25.3±9.2。表明治疗各组与对照组相比细胞凋亡数明显减少,差异有统计学用意义(P〈0.05)。联合用药组与单独给药组差异具有统计学意义(P〈0.05)。结论:黄芪、尼莫地平联合用药可以明显抑制脑缺血再灌注时神经细胞凋亡的发生,减少凋亡细胞数目,减轻脑缺血再灌注时对脑细胞的损害。

Effects of Radix Astragal and nimotop on apoptosis of neuronal cell of focal brain ischemia reperfusion injury in rats

Abstract:Objective:To study the effect of Radix Astragal,nimotop and their combination on neuronal apoptosis of focal brain ischemia reperfusion injury in rats.Methods:Sixty SD rats were divided randomly into five groups,sham operative group(0 group) was given normal dose of anesthetic,control group(the first group) received normal saline 2 ml,Radix Astragal group(the second group) received Radix Astragal injection 6 ml/kg,12 h and 30 min before surgery and every 12 h respectively given once,until after 48 h;nimotop group(the third group) to give nimotop 30 mintes before ischemia and Radix Astragal-nimotop group(the forth group) was given nimotop before ischemia and Radix Astragal injection after surgey.And there were twelve rats in each group.Experimental models were established by intraluminal occlusion of the middle cerebral artery in rats.Apoptotic cells were detected by TUNEL technique.Results:The amount of Apoptosis of neuronal cell in each group as follows 0.8±0.4,48.5±11.3,33.5±10.2,30.2±8.6,25.3±9.2.They were suggested that compared with the control group,the number of apoptotic cells in other groups were reduced evidently(P0.05),especially in Radix Astragal-nimotop group.Conclusion:Both nimotop and Radix Astragal can reduce apoptosis occurrence,but this effect can be enhanced when they are applied together,and it can evidently reduce the amount of Apoptosis of neuronal cell,and lighten the injury on neuronal cells when brain ischemia reperfusion injury occurred.So making use of insulin and nimotop after cerebral ischemia can play a role in neuroprotection.
