摘    要:目的研究四氯化碳(CCl4)对大鼠睾丸组织结构及生精细胞p53蛋白表达的影响。方法清洁级雄性Wistar大鼠36只随机分为对照组和实验组,实验组另设3个小组(1 d组、1周组和2周组),每组9只。实验组背部皮下注射体积分数60%CCl4-橄榄油溶液(0.03 mL.kg-1)。1 d组于实验第1天注射,1周组实验第1天和第4天注射,2周组实验第1天、第4天、第8天和第11天注射。对照组于实验的第1天、第4天、第8天和第11天背部皮下注射橄榄油(0.012 mL.kg-1)。实验第14天处死大鼠,取睾丸、附睾称重计算脏器系数;苏木精伊红染色(HE)和天狼猩红染色观察睾丸的组织结构和生精细胞的变化;免疫组织化学染色检测睾丸生精细胞p53蛋白的表达。结果HE和天狼猩红染色显示,对照组和1 d组大鼠睾丸基膜完整,间质偶见少量纤维,生精细胞在管内排列紧密有序,可见许多精子生成;1周组和2周组部分大鼠睾丸基膜增厚,间质纤维化明显,生精细胞排列紊乱,生精上皮层数减少,精子数目较对照组明显减少(P<0.05);睾丸和附睾脏器系数各组间比较无统计学意义(P>0.05);免疫组织化学染色显示,对照组和1 d组偶见p53蛋白阳性细胞;1周组和2周组p53蛋白阳性细胞率较对照组和1 d组明显增多,精子数目明显减少(P<0.05),2周组p53蛋白阳性细胞率较1周组明显增多,精子数目明显减少,差别均有统计学意义(P<0.05)。结论CCl4可使大鼠睾丸的组织结构发生改变,生精细胞p53蛋白表达明显增强。

Abstract:Objective To study the effects of carbon tetrachloride (CCl4 ) on changes of testicular structure and p53 albumen expression of spermatogenic cells of rats. Methods 36 clean male Wistar rats were randomly divided into experimental group (1 d group, 1 week group and 2 weeks group )and control group,9 rats in each group. Back subcutaneous injection of 60% CCl4 - olive oil solution(0.03 mL ·kg^-1 ) in experimental group. The 1 d group was injected at the first day,1 week group was injected at the first day and the fourth day,2 weeks group was injected at the first day, the fourth day, the eighth day and the eleventh day. Back subcutaneous injection of olive oil (0. 012 mL·kg^-1) was done in control group at the same time with 2 weeks group. All rats were killed at the 14th day, testis and epididymis were taken and weighted for calculate the organ coefficient. HE staining and sirius red staining were used to observe testicular structure and changes of spermatogenic cells. Immunohistoehemistry was used to detect p53 albumen expression of spermatogenic cells. Results HE and sirius red staining showed that testicular basement membrane integrity and a small amount of fiber appeared occasionally in mesenchymal; the spermatogenic cells in the tube were tight and orderly, many of spermatogenesis can be observed; interstitial fibrosis and thickening were observed in parts of testieular basement membrane of 1 week group and 2 weeks group;spermatogenic cells arranged more disordered in 1 week group and 2 weeks group than those in control groups. The number of sperm in 1 week group and 2 weeks group was lower than that in control groups ( P 〈 0.05 ) ; there were no statistic difference of the testis and epididymis organ coefficient in each group( P 〉 0.05 ). There was few p53 positive cells in the control group and 1 d group. The number of 1353 positive cell in 1 week group and 2 weeks group were much higher than that in control group and 1 day group(P 〈0.05). The number of p53 positive cell
