马钱子碱诱导人多发性骨髓瘤细胞株U266细胞凋亡与bax基因的表达

摘    要:背景:马钱子碱具有抗肿瘤作用,而其对人多发性骨髓瘤细胞生长的影响尚不明确。目的:探讨马钱子碱诱导人多发性骨髓瘤U266细胞凋亡的机制。方法:取对数生长期的U266细胞,分别用0,0.05,0.1,0.2,0.4 g/L的马钱子碱干预12,24,48 h,通过MTT法测得IC50,并以此浓度干预U266细胞,通过形态学观察、RT-PCR法检测马钱子碱对人多发性骨髓瘤U266细胞株的诱导凋亡作用。结果与结论:马钱子碱诱导U266的凋亡呈时间和浓度依赖性(P < 0.05),其作用48 h的IC50为0.16 g/L。在马钱子碱诱导的凋亡细胞中可见凋亡小体,同时,细胞中的促凋亡基因bax的表达量随马钱子碱作用时间的延长逐渐增加(P < 0.01)。说明0.4 g/L浓度范围内的马钱子碱具有诱导U266细胞凋亡的作用,这种作用具有浓度和时间依赖性,可能是通过激活bax基因途径实现的。

Abstract:BACKGROUND: Brucine has anti-tumor effect. However, its influence on growth of multiple myeloma cells is not clear. OBJECTIVE:To investigate apoptotic mechanism of brucine on human multiple myeloma.METHODS:Logarithmic growth phase of U266 cells were treated with 0, 0.05, 0.1, 0.2, and 0.4 g/L brucine for 12, 24, and 48 hours, measured IC50 by MTT, and interfere the U266 cells. The apoptotic effect of brucine on human multiple myeloma cell line-U266 was observed by morphological observation and RT-PCR.RESULTS AND CONCLUSION: Brucine induced U266 apoptosis showed a time- and concentration-dependent manner (P < 0.05). The IC50 level was 0.16 g/L at 48 hours after intervention. Apoptotic body could be found in brucine treated-apoptotic cells, meantime, the expression of pro-apoptosis gene bax was gradually increased with time prolonged (P < 0.01). It is suggest that 0.4 g/L concentration brucine induced U266 apoptosis, and the effect showed in a concentration- and time-dependent manner, which possibly through the activation of bax pathway.
