蛇床子素干预人增生性瘢痕成纤维细胞增殖及转化生长因子β1的表达

摘    要:背景:蛇床子素对体外培养人增生性瘢痕成纤维细胞增殖和细胞分泌的转化生长因子β1有抑制作用,但其具体作用机制尚待进一步研究。 目的:体外观察蛇床子素对人增生性瘢痕成纤维细胞增殖以及对细胞转化生长因子β1的影响。 方法:体外原代培养人增生性瘢痕成纤维细胞,以不同浓度的蛇床子素作用于成纤维细胞,观察细胞形态的变化,应用MTT法和生长曲线法检测蛇床子素对细胞增殖活性的影响。免疫组织化学检测细胞转化生长因子β1的表达。 结果与结论:蛇床子素能明显抑制人增生性瘢痕成纤维细胞的生长。MTT法检测的IC50为(15.2±2.0) μmol/L,可以明显下调细胞转化生长因子β1的表达(P < 0.05)。说明蛇床子素对人增生性瘢痕成纤维细胞有很强的生长抑制作用并可以下调转化生长因子β1的表达。

Effects of osthole on fibroblast proliferation and transforming growth factor beta 1 expression in hypertrophic scar tissue

Abstract:BACKGROUND:Osthole exhibits inhibitory effects on human fibroblast proliferation and transforming growth factor beta 1 (TGF-β1) expression in hypertrophic scar tissue, but the precise mechanisms remains poorly understood. OBJECTIVE:To investigate the effects of osthole on human fibroblast proliferation and TGF-β1 expression in hypertrophic scar tissue. METHODS:Human hypertrophic scar fibroblasts cells were cultured in vitro and then treated by osthole at different concentrations. The growth inhibitory effects were observed by MTT assay and cell growth curve. The expression of TGF-β1 was detected by immunohistochemistry. RESULTS AND CONCLUSION:Osthole could obviously inhibit the growth of human hypertrophic scar fibroblasts. MTT assay showed that osthole IC50 value toward hypertrophic scar fibroblasts was 15.2±2.0 μmol/L. Furthermore, the results of cell growth curve matched with the above results. Immunohistochemistry results showed that osthole could obviously inhibit TGF-β1 expression in the fibroblasts derived from hypertrophic scar tissue compared with the control group (P < 0.05). These findings suggest that osthole strongly inhibits the growth of human hypertrophic scar fibroblasts and decreases the expression of TGF-β1.
