血栓调节蛋白、细胞内黏附分子-1及氧化低密度脂蛋白内皮受体-1在糖尿病大鼠肾皮质的基因表达

摘    要:目的采用逆转录-聚合酶链反应(RT-PCR)法逆转录链脲佐菌素(STZ)诱导的糖尿病大鼠肾皮质中血栓调节蛋白(TM)、细胞内黏附分子-1(ICAM-1)及氧化低密度脂蛋白内皮受体-1(LOX-1)基因的mRNA表达,并采用半定量法测定其mRNA含量,以探讨其在糖尿病肾病的发生、发展中可能参与的机制.方法选择18只成年SD大鼠,注射STZ制成糖尿病模型,其中4只给予注射胰岛素作为治疗对照(TC)组,其余为糖尿病(DM)组;另选4只SD大鼠作为正常对照(NC)组.每4周测定尿白蛋白定量,14周取肾皮质匀浆后采用R-PCR法检测其中TM、ICAM-1及LOX-1的mRNA表达,并采用半定量法测定其含量变化.结果DM组与NC组肾皮质内TM、ICAM-1及LOX-1 mRNA含量的差异有显著性(P<0.05),与TC组的差异无显著性(P>0.05);DM组TM、ICAM-1及LOX-1的含量与尿白蛋白定量呈显著正相关(r=0.703、0.624、0.851,P<0.01、0.05和0.01).结论TM、ICAM-1及LOX-1参与了糖尿病肾病的发生、发展.

Abstract:Objective To investigate the mRNA of thromobomodulin(TM) , intercellular adhesion molecule (ICAM)-l and lectin-like oxidized low-denstity lipoprotein receptor(LOX)-1 in the kidney of streptozotocin induced diabetic rat in the roles of diabetic nephropathy. Methods 18 of 22 male Sprague-Dawley rats were injected streptozotocin to induce diabetes, 4 of them were given insulin as the therapeutic group, the rest as the diabetic group; and the other 4 constituted the control group. Urine albumin content was assayed every four weeks. Then after 14 weeks, RT-PCR was undertaken to detect the expressions of TM, ICAM-1 and LOX-1 mRNA in the renal cortex of the rats. Results TM, ICAM-1 and LOX-1 mRNA increased significantly in the kidney of diabetic group vs control group(P<0. 05), but they were not decreased in the therapeutic group receiving insulin. The urine albumin content was correlated with the increment of TM, ICAM-1 and LOX-1. Conclusion TM, ICAM-1 and LOX-1 are upregulated in diabetic kidney, they may play important roles in the development and progression of diabetic nephropathy. (Shanghai Med J, 2006, 29:309-311)
