大肠癌中survivin表达及相关性

目的研究凋亡抑制蛋白家族的新成员survivin表达变化及其与大肠癌生物学行为、bcl-2、ki-67标记指数的关系。方法应用免疫组化链霉菌抗生物素-过氧化物酶（SP）法分别检测大肠癌及其癌瘤手术切除的切缘组织survivin蛋白、bcl-2蛋白的单一及联合表达情况及其与ki-67标记指数的关系。结果（1）Survivin蛋白在大肠癌组织中的表达阳性率为63．6％，在切缘组织中的表达阳性率为0％，肠癌组织与切缘组织的表达阳性率相比差异有统计学意义（P〈0．01）。Survivin蛋白表达与患者年龄、肿瘤分化程度、肿瘤大小、部位、Dukes分期、淋巴转移无显著相关性（P〉0．05），与患者性别呈显著相关（P〈0．05）；（2）Bcl-2表达阳性与survivin蛋白表达阳性无显著相关性（P〉0．05）；（3）Survivin蛋白阳性表达组ki-67标记指数显著高于阴性组（P〈0．01）。结论Survivin基因通过调控细胞增生，参与大肠癌的发生和发展，其蛋白过表达是大肠癌发生中的一种独立事件。

Expression of survivin in colorectal cancer and correlation

Objective To study the relationship between survivin(the new member of inhibition apoptosis protein family)expression and the biological behaviors of colorectal cancer,the relationship between survivin expression and bcl-2 expression,ki-67 label index.Methods Immunohistochemical SP method was used to examine the single and joint expression of survivin,bcl-2 proteins,and the ki-67 label index in cancer tissue and marge tissue.Results(1)The positive rates of survivin expression were 63.6% in cancer tissue and 0% in marge tissue respectively.The difference between the rates was significant(P<0.01).Survivin expression didn't have significant relationship with patient age,differentiation grade,tumor size,tumor location,Duke's stage and lymph node metastasis(P>0.05),but had significant relationship with sex of colorectal cancer patients(P<0.05).(2)The bcl-2 expression didn't have significant relationship with the survivin expression(P>0.05).(3)The mean ki-67 label index in survivin positive tumors was significantly higher than that of survivin negative tumors(P<0.01).Conclusion Survivin plays a certain role in the oncogenesis and progression of colorectal cancer by up-regulating cell proliferation;the over expression of survivin is an independent event in the colorectal cancer tumorigenesis.
