摘    要:目的:观察诱导分化剂苯乙酸对胶质瘤细胞C6的诱导分化作用,并在分子水平上探讨其作用机理。方法:细胞计数和〔3H〕-TdR掺入法检测细胞增殖,流式细胞术分析细胞周期。结果:苯乙酸对细胞增殖存在剂量(25mmol/L,50mmol/L)依赖性抑制;免疫细胞化学检测CPM值降低,肿瘤细胞DNA合成减少;细胞周期分析G0/G1期所占比例下降,S期相对升高。结论:苯乙酸可阻抑细胞增殖周期,使细胞周期增殖循环中止于S期中DNA合成准备期S0期,DNA合成减少,抑制细胞增殖。其作用机理为抑制细胞周期中G1期的RNA和蛋白质合成

Abstract:Objective:To investigate phenylacetate′s function of suppressing the growth of glioma C 6 cell and inducing differentitation and find the mechanism of this effect on molecular level.Methods:Cell proliferation was evaluated Using 3H -TdR and cell numeration,using a hemocytometer.Cell proliferation cycle was tested by flow cytometry (FCM).Results:NaPA (2.5 mmol/L and 5.0 mmol/L) exerted a marked inhibitory effect on cell Proliferation. Exposure to NaPA (2.5 mmol/L and 5.0 mmol/L for 5 days) resulted in a decrease of cells in G 0/G 1 Phase as being analyzed by flow cytometry.Conclusion:NaPA could exert a marked inhibitory effect on cell proliferation and glioma malignancy becomes lower.NaPA made cell cycle distribition on G 1 phase decreased and cell cycle stopped on S 0 phase (prepare period of DNA synthesis) by indreasing RNA and protein.
