摘    要:目的:探讨亚低温对大鼠脑缺血再灌注损伤后神经细胞凋亡和Caspase—3活性的影响。方法:采用大鼠局灶性脑缺血再灌注损伤模型,大脑中动脉阻塞2h,再灌注损伤12h,用TUNEL法和荧光四肽底物(AC—DEVD-AMC)法分别检测假手术组、对照组和亚低温组的凋亡细胞百分率和Caspase—3活性水平。结果:与对照组比较,亚低温组和假手术组的凋亡细胞百分率和Caspase活性水平显著降低。结论:Caspase—3活性明显降低可能是亚低温处理大鼠脑缺血再灌注损伤后凋亡细胞百分率显著下降的分子机制之一。

Abstract:Objective: To study the effects of mild hypothermia on nervous cell apoptosis and activity of Caspase 3 after rat focal cerebral ischemia reperfusion injury. Methods: By using the model of rat focal cerebral ischemia reperfusion injury, the middle cerebral arteries (MCA) of SD rats were occluded for 2 h, and reperfused for 12 h. Using methods of TUNEL and AC DEVD AMC, the apoptotic cell rate and Caspase 3 activity level were determined in the sham operated group, the control group and the mild hypothermia group separatively. Results: the apoptotic cell rate and Caspase 3 activity level were significantly lower in the sham operated group and the mild hypothermia group than those in the control group. Conclusion: The obviously decreased activity of Caspase 3 may be one of the molecular mechanisms of significantly decreased apoptotic cell rate after rat cerebral ischemia reperfusion injury treated by the mild hypothermia.
