内皮素和一氧化氮合成酶在家兔动脉粥样硬化斑块中的表达

摘    要:用免疫组织化学和原位杂交等方法观察了内皮素和一氧化氮合成酶在家兔动脉粥样硬化斑块中的合成和表达,结果是,在动脉粥样硬化斑块内增生的平滑肌细胞和巨噬细胞中,内皮素合成较正常者增多,而一氧化氮合成酶的合成却明显减少,内皮素mRNA转录显著增加,而一氧化氮合成酶mRNA转录明显减少或消失,提示在动脉血管平滑肌细胞中,二者的平衡失调可能参与了动脉弱样硬化的发生和发展过程。

Abstract:Present test observated the synthesis and expression of endothelin(ET) and nitric oxide synthetase(NOS) in rabbit'' s atherosclerotic plaque(AP) on polypeptide and gene transfer.Histochemistry and immunohistochemistry shown that in proliferating vascular smooth muscle cell (VSMC) within AP, synthesis of ET was increased while that of NOS was significantly decreased;Meanwhile, in situ hybridization also shown that the transfer of ET mRNA was great increased while that of NOS mRNA was great decreased within 5 cases and only within 1 case,a little of NOS mRNA was expressed and the range was limited.So we believed that in aortic VSMC, un-balence of between ET and NOS may be involved in the occurrence and development of AP.
