HPLC法同时测定人血清中氯氮平与喹硫平的浓度

目的:建立同时测定人血清中氯氮平与喹硫平浓度的方法。方法:人血清样品经0.01 mol/L氢氧化钠溶液碱化后,采用正戊烷萃取。以高效液相色谱(HPLC)法进样分析,色谱柱为SB-C18,流动相为甲醇-水-四甲基乙二胺-冰醋酸(620∶380∶4.4∶3.52),流速为0.8 ml/min,柱温为40℃,紫外检测波长为254 nm,进样量为20μl,内标为2-氨基-5-氯-2′-氟二苯甲酮。结果:氯氮平、喹硫平血药浓度分别在10¹ 200、101 200、10⁷00μg/L范围内线性关系良好(r分别为0.998 5、0.996 3),定量下限均为10μg/L。氯氮平与喹硫平高、中、低3个浓度的提取回收率均>70%,日内、日间的RSD均<15%(n=5)。结论:本方法灵敏度高,操作简便、快速,结果准确,可用于人血清中氯氮平和喹硫平血药浓度测定。

Concentration Determination of Clozapinc and Quetiapine in Human Serum by HPLC

OBJECTIVE： To establish a method for simultaneous determination of clozapine and quetiapine concentration in hu- man serum. METHODS： After alkalizing by 0.01 mol/L NaOH, human serum samples were extracted by n-pentane and analyzed by HPLC. The determination was performed on SB-C18 column with mobile phase consisted of methanol-water-tetramethylethylenedi- amine-acetic acid （620 ： 380 ： 4.4 ： 3.52） at the flow rate of 0.8 ml/min. The column temperature was 40 ℃ and UV detection wave- length was set at 254 nm. Injection volume was 20 μl, and 2-amino-5-chloro-2＇ -fluoro benzophenone was used as internal standard. RESULTS： The linear range were 10-1 200 μg/L for clozapine （r=0.998 5） and 10-700 μg/L for quetiapine （r=0.996 3）, respec- tively, the quantitative lower limit were both 10 μg/L. The extraction recoveries of elozapine and quetiapine at high, middle and low concentrations were greater than 70%. The intra-day and inter-day RSD were less than 15% （n=5）. CONCLUSIONS： The method is sensitive, simple, rapid and accurate, and can be used for the determination of clozapine and quetiapine concentration in human serum.
