摘    要:目的:建立同时测定双黄连颗粒中绿原酸、木犀草苷、连翘苷和黄芩苷含量的方法。方法:采用高效液相色谱法。色谱柱为Eclipse XDB-C18,流动相为甲醇-0.4%磷酸溶液(梯度洗脱),流速为0.80 ml/min,检测波长为277 nm,柱温为30℃,进样量为10μl。结果:绿原酸、木犀草苷、连翘苷和黄芩苷检测质量浓度分别在3.9178.3、0.8478.3、0.8416.7、1.8316.7、1.8336.5、41.4736.5、41.47829.4μg/ml范围内与各自峰面积积分值呈良好的线性关系(r=0.999 8、0.999 9、0.999 8、0.999 9);精密度、稳定性、重复性试验的RSD≤1.23%;平均加样回收率分别为100.59%、101.04%、100.90%、100.76%,RSD分别为1.50%、2.05%、1.95%、1.46%(n=9)。结论:该方法简便、准确、重复性好,可用于双黄连颗粒的质量控制。

Content Determination of Chlorogenic Acid,Luteolin, Forsythin and Baicalin in Shuanghuanglian Gran- ules by HPLC

Abstract:OBJECTIVE: To establish the method for content determination of chlorogenic acid, luteolin, forsythin and ba- icalin in Shuanghuanglian granules. METHODS: HPLC method was adopted. Eclipse XDB-C18 column was used with mobile phase consisted of methanol-0.4%phosphoric acid solution (gradient elution) at the flow rate of 0.80 ml/min. The detection wavelength was set at 277 nm. The colmnn temperature was 30 % and the injection volume was 10 /al. RESULTS: The linear range were 3.91-78.3 μg/ml for chlorogenic acid(r=0.999 8), 0.84-16.7 μg/ml for luteolin(r=0.999 9), 1.83-36.5 μg/ml for forsythin(r= 0.999 8)and 41.47-829.4 μg/ml for baicalin(r=0.999 9). RSDs of precision, Stability and reproducibility tests were all lower than 1.23%. The average recoveries were 100.59% (RSD=1.50%, n=9), 101.04% (RSD=2.05%, n=9), 100.90% (RSD= 1.95%, n=9) and 100.76% (RSD=1.46%, n=9). CONCLUSIONS: The method is simple, accurate and reproducible, and can be used for the quality control of the granules.
