徐长卿6个提取部位体外抗水痘带状疱疹病毒作用研究

摘    要:目的:研究徐长卿6个提取部位体外抗水痘带状疱疹病毒(VZV)的作用。方法:以MTT法测定312.510 000.0μg/ml质量浓度范围内徐长卿6个提取部位对非洲绿猴肾(Vero)细胞的毒性;倒置显微镜下观察187.510 000.0μg/ml质量浓度范围内徐长卿6个提取部位对非洲绿猴肾(Vero)细胞的毒性;倒置显微镜下观察187.53 000.0μg/ml质量浓度范围内徐长卿6个提取部位抗VZV吸附的作用;计算半数有效浓度(EC50),以评价187.53 000.0μg/ml质量浓度范围内徐长卿6个提取部位抗VZV吸附的作用;计算半数有效浓度(EC50),以评价187.53 000.0μg/ml质量浓度范围内徐长卿6个提取部位对VZV的杀伤作用与抑制VZV在Vero细胞中的增殖作用。结果:徐长卿提取物6个部位药物质量浓度与细胞存活率之间存在线性关系;除200μg/ml徐长卿石油醚部位对VZV无吸附作用外,其余各提取部位均在相应质量浓度下对VZV吸附有一定抑制作用;徐长卿6个提取部位在相应质量浓度下对VZV有一定杀伤作用与抑制VZV在Vero细胞中的增殖作用。结论:徐长卿超临界萃取部位具有较强的体外抗VZV活性。

Abstract:OBJECTIVE:To study the antivirus effect of 6 extract parts of Cynanchum paniculatum against varicella-zoster virus(VZV)in vitro. METHODS:The toxicity of 6 extract parts of C. paniculatum(312.5-10 000.0 μg/ml)to African green monkey kidney cell(Vero)were determined by MTT assay. The absorption effects of different concentrations of 6 extract parts of C. paniculatum on VZV were observed by inverted microscope. The lethal effect of different concentrations of 6 extract parts of C. paniculatum on VZV and its inhibitory effect on the proliferation of VZV were evaluated by calculating EC50. RESULTS:There was a linear relationship between the concentrations of 6 extract parts of C. paniculatum and survival rate of cells;except for 200 μg/ml petroleum ether extract fraction from C. paniculatum,different concentrations of other parts inhibited the absorption of VZV. Different concentrations of 6 extract parts of C. paniculatum showed lethal effect on VZV and inhibitory effect on the proliferation of VZV.CONCLUSIONS:CO2-SFE part of C. paniculatum had obviously direct inhibitory effects on VZV in vitro.
