消氮颗粒对腺嘌呤所致慢性肾衰模型大鼠的保护作用

摘    要:目的:研究消氮颗粒对腺嘌呤所致慢性肾衰模型大鼠的保护作用。方法:灌胃腺嘌呤(200 mg/kg),每天1次,连续30 d,以复制大鼠慢性肾衰模型。60只Wistar大鼠随机均分为正常对照(等容纯化水)组、模型(等容纯化水)组、尿毒清颗粒(2.088 g/kg)组与消氮颗粒高、中、低剂量(3.956、1.977、0.989 g/kg)组,灌胃给药,每天1次,连续30 d。测定大鼠血肌酐(Scr)、尿素氮(BUN)、尿酸(UA)、电解质钾(K+)、钠(Na+)、钙(Ca2+)、磷(P3+)]、尿蛋白(PRO)、中性粒细胞(LEU)、尿比重(SG),光镜观察大鼠肾脏病理学变化。结果:与正常对照组比较,模型组大鼠血清Scr、BUN、UA、P3+水平升高,尿液PRO、LEU水平升高,SG降低,差异有统计学意义(P<0.01);大鼠肾脏呈灰白色,系膜区明显增宽,系膜基质明显增多,小管间质可见大量炎细胞浸润,血管壁增厚;与模型组比较,消氮颗粒高、中、低剂量组大鼠血清Scr、BUN、UA、P3+水平降低,尿液中PRO、LEU水平降低,SG升高,差异有统计学意义(P<0.01或P<0.05);大鼠肾组织病理情况有一定改善。结论:消氮颗粒对腺嘌呤所致慢性肾衰大鼠有改善作用。

Protective Effects of Xiaodan Granules on Renal Function of Rats with Chronic Renal Failure Induced by Adenine

Abstract:OBJECTIVE:To study the protective effects of Xiaodan granules on renal function of rats with chronic renal failure(CRF)induced by adenine. METHORDS:60 Wistar rats were randomly divided into normal control group(constant volume of normal saline),model group(constant volume of normal saline),Niaoduqing granules group(2.088 g/kg)and Xiaodan granules high-dose,medium-dose and low-dose groups(3.956,1.977,0.989 g/kg). They were given relevant medicines intragastrically,once a day,for consecutive 30 days. The renal failure model of rats was induced by intragastric administration of adenine(200 mg/kg),once a day,for consecutive 30 days. The serum creatinine(Scr),blood urea nitrogen(BUN),uric acid(UA),electrolyte K+,Na+,Ca2+,P3+,proteinuria(PRO),neutrophils(LEU)and urine specific gravity(SG)were determined,and renal pathology was observed under light microscope. RESULTS:Compared with normal control group,the serum contents of Scr,BUN,UA and P3+,and the contents of PRO and LEU in urine were increased in model group,while SG level was decreased;there was statistical significance(P〈0.01). Renal tissue of rats was gray white,and intraglomerular mesangial region broadened significantly. The intraglomerular mesangial matrix increased significantly,and a large number of inflammatory cell infiltration and thickening of blood vessels were found in renal tubules and interstitium. Compared with model group,the serum contents of Scr,BUN,UA and P3+,the contents of PRO and LEU in urine were decreased in Xiaodan granules high-dose,medium-dose and low-dose groups,while SG level was increased;there was statistical significance(P〈0.01,P〈0.05). The symptom of renal pathology had been improved to certain extent. CONCLUSIONS:Xiaodan granules have definitely therapeutic effect on chronic renal failure of rats caused by adenine.
