摘    要:目的研究转化生长因子β1(TGF—β1)对大鼠肝星状细胞(HSC)表达β-连环蛋白(β-catenin)的影响。方法采用逆转录聚合酶链反应(RT—PCR)比较经不同浓度的TGF-β1刺激不同时间后HSC表达β-catenin、smad3和α-SMA mRNA的变化,并比较三者之间的关系。结果1ng/ml TGF—β1刺激2h后,HSC表达β—catenin mRNA、smad3 mRNA和α—SMA mRNA量最大,β—catenin mRNA表达与两者均具有明显的相关关系(r=0.947,P〈0.01;r=0.950,P〈0.01)。结论β—catenin在TGF—β1活化HSC的过程中发挥重要作用。

Abstract:Objective To observe the effects of transforming growth factor beta 1 (TGF-heta 1 ) on expression of beta- catenin in hepatic stellate ceils during hepatic fibrosis. Methods RT PCR was used to observe the expression of beta catenin mRNA, smad3 mRNA and alpha-SMA mRNA in the hepatic stellate cells after being stimulated by different concentration of TGF-beta1 with different time. The correlation was also analyzed. Results The expression of beta- catenin mRNA, smad3 mRNA and apha-SMA mRNA in the hepatic stellate cells all reached to the maximal after being stimulated by 1 ng/ml TGF-betal for 2 hours. There were positive correlations among the three genes expressions (r = 0. 947, P〈0.01 ; r = 0. 950, P〈0.01 ). Conclusion Beta-catenin plays an important role in the activation of hepatic stellate cells by TGF beta 1.
