摘    要:目的:探究Twist调控对卵巢癌细胞侵袭和转移的影响及其机制。方法:从我院2016年3月至2017年3月间确诊的卵巢癌患者中选择75例作为研究组,其中早期14例,中期42例,晚期19例;另随机选择同期来我院体检的健康人75例作为对照组。收集两组患者的卵巢组织,用免疫组化的方法检测Twist、N-cadherin与E-cadherin和MMP-9蛋白的表达情况,用RT-PCR法检测Twist、N-cadherin与E-cadherin、细胞凋亡相关基因(Bcl-2、Bax)mRNA表达水平。结果:研究组患者卵巢组织中的Twist、MMP-9蛋白阳性表达率显著高于对照组患者(P<0.05);研究组患者卵巢组织中N-cadherin蛋白阳性表达率显著高于对照组患者(P<0.05),而对照组患者E-cadherin蛋白阳性表达率显著高于研究组患者(P<0.05);研究组患者卵巢组织中Twist、N-cadherin mRNA表达水平显著高于对照组患者(P<0.05),对照组患者E-cadherin mRNA表达水平显著高于研究组患者(P<0.05);研究组患者卵巢组织中Bcl-2 mRNA表达水平显著高于对照组患者(P<0.05),对照组患者Bax mRNA表达水平显著高于研究组患者(P<0.05)。 结论:Twist在卵巢癌的侵袭与转移中发挥了重要的作用,其参与了上皮间质转化的过程。

Abstract:Objective:To investigate the effect of Twist regulation on invasion and metastasis of ovarian cancer cells and its mechanism.Methods:From March 2016 to March 2017,75 patients with ovarian cancer were randomly selected as the study group,and 75 healthy people were randomly selected as the control group,14 cases in early stage,42 cases in middle stage and 19 cases in late stage.The expression of Twist,N-cadherin,E-cadherin and MMP-9 proteins in ovarian tissues of the two groups were detected by immunohistochemistry.The expression levels of Twist,N-cadherin,E-cadherin,Bcl-2 and Bax mRNA were detected by RT-PCR.Results:The positive expression rates of Twist and MMP-9 protein in ovarian tissues of the study group were significantly higher than those of the control group (P<0.05),and the positive expression rates of N-cadherin protein in ovaries of the study group were significantly higher than those of the control group (P<0.05),while the positive expression rates of E-cadherin protein in the control group were significantly higher than those of the study group (P<0.05).The expression levels of Twist and N-cadherin mRNA in the ovaries of the study group were significantly higher than those of the control group (P<0.05).The expression level of E-cadherin mRNA in the control group was significantly higher than that of the study group (P<0.05),and the expression level of Bcl-2 mRNA in the ovaries of the study group was significantly higher than that of the control group (P<0.05).The Bax mRNA expression level was significantly higher in the control group than that in the study group (P<0.05).Conclusion:Twist plays an important role in the invasion and metastasis of ovarian cancer,and is involved in the process of epithelial-mesenchymal transition.
