加味补阳还五汤在大鼠视神经损伤修复中的作用研究

摘    要:目的 探讨加味补阳还五汤在视神经挫伤大鼠损伤修复中的具体作用机制,为临床提供实验依据。方法 30只SD大鼠随机分为药物组、生理盐水组及空白对照组,前两组大鼠右眼建立视神经损伤模型后分别给予加味补阳还五汤、生理盐水灌胃,空白对照组不做处理。分别于造模前及造模后1 d、1周、2周、4周、6周,各组均行闪光视觉诱发电位观察P波振幅和潜伏期的变化,HE染色观察视网膜组织形态学改变,免疫组织化学染色检测视网膜Caspase-3蛋白表达的变化。结果 闪光视觉诱发电位结果显示,造模后4周、6周,药物组P波潜伏期明显缩短,P波振幅趋于平稳,与生理盐水组相比差异均有统计学意义(均为P<0.05)。HE染色结果显示,空白对照组视网膜组织结构层次清晰,药物组和生理盐水组造模后可见视网膜组织结构紊乱,明显水肿;随时间推移与生理盐水组相比,药物组的视网膜组织形态逐渐恢复,视网膜神经节细胞形态逐渐接近空白对照组。免疫组织化学染色结果显示,生理盐水组及药物组均在造模后1 d Caspase-3蛋白开始表达,造模后1周达到峰值,而后逐渐减少;与生理盐水组相比,造模后2周、4周、6周药物组Caspase-3蛋白表达均显著降低(均为P<0.05)。结论 加味补阳还五汤在大鼠视神经损伤后能改善视神经的损伤,促进视神经修复。

Abstract:Objective To explore the specific therapeutic effect of modified Buyang Huanwu Decoction on optic nerve repair in rats and to provide a reliable experiment basis for clinical practice.Methods A total of 30 healthy SD rats were randomly divided into decoction group,normal saline group and blank control group.Rats in the decoction group and normal saline group were given intragastric administration with modified Buyang Huanwu decoction and normal saline accordingly after the optic nerve injury modeling for their right eyes were established,and rats of blank control group were fed normally without any treatment.The changes in amplitude and latency of flash visual evoked potential (F-VEP) blank P wave of each group were observed before model establishment and 1 day,1 week,2 weeks,4 weeks and 6 weeks after model establishment.HE stanning method was adopted to observe changes in retinal morphology under light microscope and immunohistochemical stanning was used to detect expression of protein Caspase-3 in retina.Results Results of F-VEP indicated that,4 weeks and 6 weeks after model establishment,the latency of P wave of the decoction group was significantly shortened and amplitude of P wave inclined to be stable,and the difference was statistically significant compared with normal saline group (all P<0.05).HE stanning indicated that,retinal structure of rats in blank control group was clear while retinal structures in the decoction group and normal saline group after model establishment were disordered with obvious visible edema.Compared with normal saline group,the morphology of retinal tissue of rats in the decoction group gradually recovered over time and morphology of their ganglion cells gradually approached similar to that of the blank control group.Immunohistochemical stanning showed that Caspace-3 protein of normal saline group and decoction group began to express on the first day after model establishment and reached the peak 1 week later and then gradually decreased;compared with normal saline group,the expression of Caspace-3 protein in the decoction group were decreased 2 weeks,4 weeks and 6 weeks after model establishment,with statistically significant (all P<0.05).Conclusion The modified Buyang Huanwu decoction can help improve the optic nerve injuries of rats and facilitate their optic nerve repair.
