硒对铅致TK6细胞遗传损伤的拮抗作用

摘    要:目的 研究亚硒酸钠对铅致TK6细胞遗传毒性损伤的拮抗作用。方法 培养TK6细胞,调整细胞密度为1×106/mL,试验分3组,正常对照组、铅染毒组和亚硒酸钠干预组,采用免疫荧光染色检测细胞DNA双链断裂损伤生物标志物γH2AX、ELISA试剂盒检测细胞DNA氧化损伤生物标志物8-羟基脱氧鸟苷(8-OHdG)和染色体畸变试验检测细胞染色体损伤情况。结果 铅染毒后,TK6细胞内γH2AX含量、8-OHdG水平和染色体畸变数明显增高,与正常对照组比较差异有统计学意义(P<0.01);0.01 μg/mL的亚硒酸钠干预后,细胞内γH2AX含量和染色体畸变数下降,与铅染毒组比较差异有统计学意义(P<0.01),细胞内8-OHdG水平明显下降,与铅染毒组比较差异有统计学意义(P<0.01),而与正常对照组比较差异无统计学意义(P>0.05)。结论 铅对TK6细胞有遗传毒性损伤作用,0.01 μg/mL的亚硒酸钠具有拮抗铅对TK6细胞遗传毒性损伤的作用。

Abstract:Objective To study the antagonistic effect of sodium selenite on the genetic toxicity induced by lead in TK6 cells. Methods TK6 cells were cultured and the cell density was adjusted to 1×106/ml. The cells were divided into 3 groups: a normal control group, a lead exposed group, and a sodium selenite intervention group. Immunofluorescence staining was used to detect DNA double strand break biomarker γH 2AX, ELISA kit was used to detect DNA oxidative damage biomarker 8-hydroxydeoxyguanosine (8-OHdG), and the chromosome aberration test was used to detect cell chromosome damage. Results After lead exposure, the content of γH2AX, the level of 8-OHdG, and the number of chromosome aberrations in TK6 cells were significantly higher than those in the control group (P<0.01). After treated with sodium selenite at 0.01μg/ml, the content of γH2AX and the number of chromosome aberrations decreased significantly, which was significantly different from that of lead exposure group (P<0.01). and the intracellular 8-OHdG level decreased significantly (P<0.01), which was significantly different from that of the lead exposure group (P<0.01), but had no significant difference from that of the normal control group (P>0.05). Conclusion Lead has genotoxic damage on TK6 cells, and 0.01μg/ml sodium selenite can antagonize the genetic damage induced by lead in TK6 cells.
