微小RNA-106a在膀胱癌组织中的表达及其对膀胱癌T24细胞增殖、侵袭作用的影响

摘    要:目的:观察微小RNA-106a(miR-106a)在膀胱癌组织中的表达及其对膀胱癌T24细胞增殖、侵袭作用的影响。方法:提取35例膀胱癌组织及正常膀胱组织中的总RNA,实时定量RT-PCR法检测miR-106a在膀胱癌组织及正常膀胱组织中的表达量;培养膀胱癌T24细胞,分成实验组及对照组,实验组转染miR-106a mimics,对照组转染空白脂质体,噻唑蓝(MTT)法检测转染miR-106a mimics后对T24细胞增殖的影响,流式细胞仪检测T24细胞周期变化,Transwell侵袭小室检测T24细胞侵袭能力。结果:膀胱癌组织中miR-106a的表达量显著低于正常膀胱组织。膀胱癌T24细胞转染miR-106a mimics后,T24细胞增殖能力降低,细胞周期停滞于G_1期,Transwell侵袭小室检测提示T24细胞侵袭能力降低。结论:miR-106a的表达量在膀胱癌组织中减少,miR-106a过表达抑制膀胱癌T24细胞的增殖及侵袭能力。

Abstract:Objective:To observe the expression of microRNA-106a(miR-106a) in bladder cancer and its effect on the proliferation and invasion of bladder cancer T24 cells.Methods:The total RNA was extracted from 35 cases of bladder cancer and normal tissues.The expression of miR-106a in bladder cancer tissues and normal tissues was detected by real-time quantitative RT-PCR.The bladder cancer T24 cells were cultured and divided into experimental group and control group.The experimental group was transfected with miR-106a mimics,and the control group was transfected with only lipofectamine.The proliferation of T24 cells was detected by MTT assay.The cell cycle of T24 cells was detected by flow cytometry.The invasion of T24 cells was detected by Transwell invasive chamber.Results:The expression of miR-106a in bladder cancer tissues was significantly lower than that in normal bladder tissues.Transfection of miR-106a mimics decreased the proliferation of T24 cells and arrested the cell cycle in G1 phase.Transwell invasive assay revealed that the invasive ability of T24 cells was decreased.Conclusion:The expression of miR-106a is decreased in bladder cancer tissues,and overexpression of miR-106a inhibited the proliferation and invasion of T24 cells.
