摘    要:目的  研究苯酚提取法对A 群脑膜炎球菌多糖料液中细菌内毒素含量的影响。方法  用苯酚对A 群脑膜炎球菌多糖料液进行提取,比较粗制多糖不同溶解倍数、苯酚提取过程中采用不同搅拌转数、提取次数、提取温度等因素对多糖细菌内毒素含量的影响。  结果  粗制多糖采用4.5%醋酸钠溶液1∶100(质量体积比)溶解后进行苯酚提取的多糖细菌内毒素含量最低。同时,综合考虑多糖料液中蛋白含量、多糖回收率的检定结果,确定用苯酚进行提取的最佳条件为搅拌转数为400 转/min,提取次数为3次,温度为13~15 ℃,可有效降低多糖料液中细菌内毒素的含量。  结论  苯酚提取法可明显降低A 群脑膜炎球菌粗制多糖料液中的细菌内毒素含量。

Effect of phenol extraction on endotoxin content in group A meningococcal polysaccharide

Abstract:Objective   To investigate the effect of phenol extraction on endotoxin content in group A meningococcal polysaccharide solution. Methods  The group A meningococcal polysaccharides were extracted with phenol. The effects of different dissolution ratios, speed of agitator, phenol extraction time, and phenol extraction temperature on the endotoxin content of polysaccharides were compared. Results  When the crude polysaccharide is dissolved by 4.5% sodium acetate solution 1∶100(mass to volume ratio) before extracted by phenol, the content of endotoxin in polysaccharides was significantly lower than that of the other two groups. Taking into account the protein content and polysaccharide recovery rate, the phenol extraction speed of 400 r/min, three times extraction, the temperature of 13-15 ℃ were choosen.  Conclusion  The phenol extraction method can significantly reduce the endotoxin contentof group A meningococcal polysaccharide.
