摘    要:目的:研究小白菊内酯对人甲状腺癌BCPAP细胞的增殖以及自噬水平的影响。方法:通过MTS法检测不同药物浓度的小白菊内酯对BCPAP细胞增殖的作用。倒置显微镜观察不同给药浓度时细胞的形态变化。免疫荧光技术检测LC3的表达情况。Western Blot法检测自噬相关蛋白LC3Ⅱ与Beclin1的变化。结果:小白菊内酯可以抑制BCPAP的增殖,呈时间与剂量依赖性。随着给药浓度的增加,细胞数量逐渐减少,且形态逐渐改变,细胞皱缩、变圆、透亮,能够反映细胞LC3表达的红光逐渐增强。 Western Blot结果显示,LC3Ⅱ与Beclin1的表达均随着给药浓度的增加而增加。结论:小白菊内酯可以提高甲状腺癌BCPAP细胞的自噬水平,且显著抑制细胞的增殖,呈时间与剂量依赖性。

Abstract:Objective:To explore the effects of parthenolide on proliferation and autophagy of human thyroid carcinoma cell line BCPAP.Methods:BCPAP cell proliferation was detected by MTS assay after the cells were treated with different doses of parthenolide.The morphological changes of cells were observed by inverted microscope.The expression of LC3 was monitored by immunofluorescence technique.The expression of autophagy-related proteins including LC3II and Beclin1 was detected by Western Blot.Results:Parthenolide can inhibit the proliferation of BCPAP cells in a time-and dose-dependent manner.With the increase of concentration of the drug,the number of cells gradually decreased,and the morphology of the cells gradually changed.The cells shrunk,were rounded,translucent,and the red light which reflected the expression of LC3 in the cells gradually increased.Western Blot results showed that the expression of LC3II and Beclin1 increased with increasing concentration of parthenolide.Conclusion:Parthenolide can increase the level of autophagy in thyroid carcinoma BCPAP cells,and significantly inhibit cell proliferation in a time-and dose-dependent manner.
