摘    要:目的建立测定胖血藤中大黄素和大黄素甲醚含量的反相高效液相色谱法。方法采用DiamonsilCts柱(250mm×4.6mm,5txm),以甲醇-0.1%磷酸溶液(80:20)为流动相,流速为1.0mL/min,检测波长为254nm,柱温为30℃。结果大黄素、大黄素甲醚的进样量分别在0.1000~1.6000μg(r=0.9999)和0.03125~0.2500txg(r=0.9997)范围内与峰面积呈良好线性关系,平均回收率分别为99.07%(RSD=1.04%,n=6)和100.05%(RSD:1.54%,n=6)。结论该方法简便、准确、可靠,可用于胖血藤的质量控制。

Abstract:Objective To develop a reversed phase high performance liquid chromatography(RP- HPLC) method for determining emodin and physcion in Radix Polygoni cynanchoidis. Methods The Diamonsil Cts column(250 mm x4. 6 mm, 5 p~m) was adopted with the mobile phase of methanol-0. 1% phosphoric acid(80:20) at the flow rate of 1.0 mL/min. The detection wavelength was set at 254 nm and the column temperature was 30 ~C. Results The linear ranges were 0. 100 0- 1. 600 0 p~g(r=0. 999 9) for emodin and 0.031 25-0.250 0 p~g(r=0.999 9) for physcion. The average recovery rates of emodin and physcion in Radix Polygoni cynanchoidis were 99.07% (RSD=l.04% , n=6) and 100.05% (RSD=l.54% , n=6) respectively. Conclusion The method is simple, accurate and reliable, which can be used to be the quality control of Radix Polygoni cynanchoidis.
