硼替佐米联合阿糖胞苷诱导对K562细胞株PTPROt基因的影响

摘    要:目的 研究硼替佐米及阿糖胞苷序贯及联合用药对K562细胞的凋亡率的影响,同时观察硼替佐米是否经由PTPROt基因的表达上调促细胞凋亡.方法 以慢性粒细胞白血病(CML)细胞的K562细胞为研究对象.不同用药组分别用药48 h,细胞选用噻唑蓝(MTT)法检测细胞增殖抑制率,采用流式细胞术观察细胞抑制率与凋亡率情况,同时观察PTPROt基因表达情况.结果 联合应用硼替佐米及阿糖胞苷组与分别应用硼替佐米及阿糖胞苷组两两相比差异有统计学意义(P<0.05).联合用药使细胞凋亡率增加,序贯先应用阿糖胞苷后硼替佐米的细胞凋亡率最高,与单用硼替佐米及阿糖胞苷组两两相比差异有统计学意义(P<0.05).应用硼替佐米后PTPROt基因表达明显上调.结论 联合硼替佐米及阿糖胞苷,对白血病细胞株K562细胞有协同增强肿瘤细胞凋亡作用,其作用机制可能与上调PTPROt基因表达相关.

Abstract:Objective To study the impact of sequential and combination medication of bortezomib and Ara-e on the apoptosis rate of K562 cells and to simuhaneously observe whether bortezomib promote apoptosis via the upregulation of the PTPROt gene expres- sion. Methods The K562 cell line in chronic myelogenous leukemia(CML) was taken as the research subjects. Ditt~rent medication groups used drugs for 48 h,The MTF assay was adopted to detect the inhibition rate of the cell prolit~ration in each group,the apopto- sis rate was detected by the flow eytometlT,the inhibition rate and the apoptosis rate were observed,at the same time the PTPROt geue expression after medication in each group was observed. Results The comparison in pairs of bortezomib and Ara- e eombinalion group with single bortezomib and Ara-c respective application had statistical significance( P 〈 O. 05).The combination medication increased the apoptosis rate,while the sequential medication of first Ara-e and then bortezomib had the highest apoptosis rate,the in pairs com- parison with single bortezomib and Ara- c application showed statistical significance (P 〈 O. 05). The PTPROt gene expression was significantly up- regulated by bortezomib application. Conclusion Bortezomib c.nmbined with Ara- c has the synergistically enhanced ef- teet on the tumor apoptosis in leukemia cell line K562 cells,its mechanism may be related with upregulating PTPROt gene expression
