复方土茯苓颗粒对大鼠肝细胞XDH-mRNA表达和XO活性的影响

摘    要:目的观察复方土茯苓颗粒含药血清对大鼠肝细胞高尿酸血症(HUA)模型尿酸(UA)、黄嘌呤氧化酶(XO)活性及肝脏中黄嘌呤氢化酶(XDH)mRNA转录水平的抑制作用。方法体外培养大鼠BRL 3A肝细胞,制备复方土茯苓颗粒中药血清,将细胞分成6组,即空白对照组、转染对照组、空白大鼠血清组、中药血清低、中、高剂量组(体积分数为5%、10%、20%),将pENTER-XDH重组质粒转染到空白组以外的BRL细胞,复制成肝细胞HUA模型,并将中药血清加入至相应组别。用qRT-PCR方法检测XDH-mRNA的转录水平,并进行UA分泌水平及XO活性检测。结果与空白对照组相比,转染对照组的XDH-mRNA转录水平明显升高,差异有统计学意义;与空白大鼠血清组相比,中药高剂量组XDH-mRNA的转录水平显著降低,并具有统计学意义;空白大鼠血清组的尿酸水平明显高于空白对照组和转染对照组,差异有统计学意义;各中药血清组与空白大鼠血清组相比,中药血清中剂量组与高剂量组的UA及XO活性要相对更低,差异有统计学意义。结论复方土茯苓颗粒可能通过降低XDH-mRNA和XO的活性来达到降尿酸的作用。

Abstract:Objective To observe the inhibition effect of Chinese herb containing serum of compound Tufuling Granules on level of uric acid(UA),xanthine oxidase(XO) activity and the expression of xanthine hydrogenase(XDH) mRNA in hyperuricemia model of rat liver cells. Methods BRL 3 A cells were cultured in vitro. The Chinese herb containing serum of compound Tufuling Granules was prepared. The BRL 3 A cells were divided into six groups:Blank Control Group(BCG),Transfection Control Group(TCG),Blank Serum Group(BSG),Low Dose(LDG),Middle Dose(MDG),and High Dose of Herb Serum Group(HDG). The pENTER-XDH plasmid was transfected to all group cells except the BCG. Blank rat serum was added to BSG,and the different concentrations of Chinese herb containing serum were added to correspondent groups. Then the XDH gene expression,UA level,and XO activity were detected. Results Compared with the BCG, the XDH-mRNA expression level of the TCG was significantly increased. The expression level of XDH-mRNA was significantly lower in the HDG compared with the BSG. The UA level in the BSG was significantly higher than that in the BCG and the TCG. Compared with the BSG,the UA and XO activities of the MDG and HDG were relatively lower with statistically significant differences.Conclusion Compound Tufuling Granules can reduce UA through reducing the XDH-mRNA expression and the activity of XO.
