复方野菊花提取物对高脂高糖饮酒致肝旺痰阻型高血压大鼠的作用研究

摘    要:目的观察复方野菊花提取物对高脂高糖饮酒致肝旺痰阻型高血压大鼠的作用。方法采用高脂高糖饲料喂养复合梯度饮酒制备肝旺痰阻型高血压大鼠模型,模型复制成功后灌胃给予复方野菊花提取物连续6周。末次给药后,观察大鼠性情变化,测定面温、抓力、眩晕时间、血压;检测血清中C反应蛋白(CRP)、内皮素-1(ET-1)、一氧化氮(NO)、丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)水平;HE染色观察大鼠胸主动脉组织形态学,Masson染色观察胸主动脉和肝脏纤维化情况;免疫组化法检测胸主动脉中细胞间黏附分子-1(ICAM-1)表达。结果复方野菊花提取物能改善高血压大鼠性情,降低面温、眩晕时间、收缩压(SBP)、舒张压(DBP)和平均动脉压(MBP);降低血清中CRP、ET-1、ALT和AST水平,升高NO含量;抑制胸主动脉中膜增厚及内皮细胞脱落,并改善胸主动脉和肝脏中胶原纤维沉积;抑制胸主动脉中ICAM-1的表达。结论复方野菊花提取物能够改善肝旺痰阻型高血压大鼠眩晕、面部烘热、狂躁易怒等证候,降低血压并能改善胸主动脉和肝脏病变;其降压机制可能与抑制血管内皮炎症损伤,平衡血管内皮舒缩因子NO/ET-1有关。

Effect of Compound Dendranthema indicum Extract on Hypertension Rats with Liver Prosperity and Phlegm Obstruction Syndrome Induced by High Glucose and High Fat Compound Alcohol

Abstract:Objective To observe the effect of compound Dendranthema indicum extract on hypertension rats with liver prosperity and phlegm obstruction syndrome induced by high glucose and high fat compound alcohol. Methods The rats were fed with high glucose and high fat compound alcohol diet to prepare the hypertension model of liver prosperity and phlegm obstruction syndrome type,and the compound Dendranthema indicum extract was given for 6 weeks after the model was successfully established. After the last administration, the changes of the rats’ temperament were observed and the surface temperature, grasping force, vertigo time and blood pressure were measured. The serum levels of C-Reactive protein (CRP),Endothelin 1 (ET-1),Nitric Oxide (NO),Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST) were measured, and the thoracic aorta tissue of rats was observed by HE staining. Morphology and Masson staining were used to observe the fibrosis of the thoracic aorta and liver. Immunohistochemical staining was used to detect the expression of intercellular cell adhesion molecule-1 (ICAM-1) in the thoracic aorta. Results The compound Dendranthema indicum extract can improve the temperament of hypertensive rats, reduce the surface temperature, vertigo time, systolic blood pressure (SBP),diastolic blood pressure (DBP) and mean blood pressure (MBP),reduce the levels of CRP,ET-1,ALT and AST in the serum,increase the content of NO,inhibit the thickening of the middle membrane of the thoracic aorta and the endothelial cells falling off,and improve the blood pressure;collagen fibers deposition in the thoracic aorta and liver,and the expression of ICAM-1 in the thoracic aorta were also inhibited. Conclusion The compound Dendranthema indicum extract can improve the syndrome of vertigo,facial heat and manic irritability in rats with liver prosperity and phlegm obstruction syndrome type hypertension, reduce blood pressure and improve the pathological changes of the thoracic aorta and liver,which may be related to the inhibition of vascular endothelial inflammation and the balance of vascular endothelial vasoconstriction factor NO/ET-1.
