高血压大鼠脑缺血再灌注损伤的观察

摘    要:目的:观察大鼠脑缺血再灌注损伤。方法:选SD 大鼠,制备肾血管性高血压大鼠(RHR),以线栓法复制大脑中动脉梗阻(MCAO) 3 h 后再灌注21 h(A 组);MCAO6 h 后再灌注18 h(B组);MCAO24 h(C组)。于MCAO 后6 h 及24 h 进行神经功能障碍评分,TTC染色后以图像分析技术测脑梗塞体积。结果:A 组大部分动物无神经功能障碍及梗死灶出现;B、C两组均全部出现不同程度神经功能障碍及梗死灶,且B组较C组更严重,具有显著性差异(P< 0.01)。结论:高血压大鼠线栓法脑缺血6 h 后再灌注18 h 可引起显著的再灌注损伤

Abstract:Objective: To study the reperfusion injury in rats with cerebral ischemia. Method: Renovascular hypertension was induced in Sprague Dewley(SD)rats for the study. Middle cerebral artery occlusion (MCAO) was done for 3 hours with reperfusion for 21 hours in Group A, MCAO for 6 hours with reperfusion for 18 hours in Group B and MCAO for 24 hours with no reperfusion in Group C respectively. Neurological deficit evaluation was then carried out at 6 hours and 24 hours after MCAO and cerebral infarction volume was examined with computer imaging techniques after TTC staining. Results: Most rats in Group A had no neurological deficit and/or cerebral infarction while Groups B and C had these problems to various degrees. Group B showed even more serious neurological deficit and greater cerebral infarction volume than Group C(P<0.01). Conclusion: Significant reperfusion injury may develop in rats with renovascular hypertension at 6 hours after cerebral ischemia with reperfusion for 18 hours afteh MCAO.
