Simultaneous determination of fourteen components in Fufang Xiling Jiedu capsules by UPLC-Q-TOF-MS

Objective:To develop an ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometric(UPLC-Q-TOF-MS)method for the determination of fourteen components in Fufang Xiling Jiedu capsules.Methods:The analysis was performed on an Agilent Advance Bio chromatographic column(250 mm×2.1 mm,2.7μm)with the mixture of 0.1% formic acid-water-acetonitrile as mobile phase,and the flow rate was0.4 mL·min^-1.The mass spectrometry was performed under negative ion mode and the fragment set at 175 V.The injection volume was 2.0 uL.Results:Good linearity under the optimal chromatographic conditions were obtained,with r2 values greater than 0.993.And the method validated in terms of good accuracy,repeatability and stability.The average recoveries(n=6)were between 97%-103% with RSDs between 0.61%-1.24%.Fourteen compounds including 3 flavonoids,9 phenolic acids,1 triterpene and 1 lignan were rapidly analyzed within 31 minutes with the established analytical method.The results showed that the highest content in Fufang Xiling Jiedu capsules was glycyrrhizic,with the average content of 9.976 mg·g^-1,followed by chlorogenic acid and its isomers,flavonoids and lignans.Conclusion:The method is simple,accurate,and reliable to determine the fourteen contents in Fufang Xiling Jiedu capsules.
