大鼠损伤组织中VEGF bFGF表达变化的免疫组化研究

摘    要:目的 :探讨大鼠锐器损伤后组织中血管内皮细胞生长因子 (VEGF)和碱性成纤维细胞生长因子 (bFGF)表达的动态变化。方法 :在大鼠背侧切开皮肤全层建立大鼠模型 ,不同时间 (0 .5、1、3、6、12、2 4、4 8、72、96h)分批处死大鼠。应用免疫组化ABC法检测VEGF、bFGF的表达 ,并用图像分析系统分析其表达情况。结果 :VEGF在损伤后12h开始表达 ,72h表达最强 ,与正常对照大鼠皮肤组织比较 ,差异有显著性 (P <0 .0 1) ,96h以后开始下降。bFGF在损伤后 1h开始表达 ,4 8h表达最强 ,与正常对照大鼠皮肤组织比较 ,差异也有显著性 (P <0 .0 5 ) ,72h后开始下降。结论 :VEGF和bFGF可望在法医学损伤时间推断中作为一种客观指标 ,但bFGF在损伤组织中表达早于VEGF ,持续的时间较VEGF长 ,因此bFGF在法医损伤时间推断中有着重要作用。

Abstract:Objective: To investigate the expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) during skin wound healing in rat. Methods: After a full-thickness incision was made on the dorsal skin, the rat was killed 0.5,1,3,6,12,24,48,72,96 h respectively after incision and the wound was excised. The expression of VEGF and bFGF in the wound tissue were detected by immunohistochemical techniques and MIAS image analysis system. Results: The increase of VEGF was observed at 12 h after injury. The strongest increase was at 72 h and it declined at 96 h. Compared with normal tissue, it showed obvious difference from 72 h (P<0.01). But the expression of bFGF was increased at 1 h, reached peak at 48 h and declined at 72 h. Compared with normal tissue, it showed obvious difference from 24 h (P<0.05). Conclusion: Time-dependent expression of VEGF and bFGF factors suggested that they would be useful markers for the determination of wound age. However, bFGF should be superior to VEGF because of its earlier expression and more persistent expression in the wound tissue.
