摘    要:目的 目的 观察钩吻素子对小鼠自主行为、协调平衡和巴比妥阈下催眠等中枢神经系统作用的影响. 方法 小鼠灌胃给予钩吻素子2,10,50 mg/kg,分别观察其在2h内的自主行为活动和协调平衡运动能力的影响,以及对阈下剂量戊巴比妥钠小鼠的催眠协同作用. 结果 50 mg/kg钩吻素子增加小鼠的自主行为活动(P<0.05),10 mg/kg钩吻素子促进阈下剂量戊巴比妥钠小鼠的睡眠作用(P<0.05),钩吻素子对小鼠协调平衡运动能力无影响. 结论 钩吻素子在治疗范围的较高剂量下能引起小鼠中枢神经系统的抑制作用,超过治疗剂量范围的一定时间内则产生兴奋作用.

Abstract:Objective To observe the effect of Koumine on the function of central nervous system (CNS) in mice with respect to spontaneous locomotor,coordinated locomotor and barbital-induced hypnosis at a sub-threshold dose.Methods The mice were intragastrically administrated with 2,10 and 50 mg/kg Koumine.Two hours after administration,the effect of Koumine on spontaneous locomotor activity,coordinated locomotor ability and pentobarbital sodium-induced hypnosis was evaluated by activity meter test,rotarod test,barbital interaction test,respectively.Results Koumine at the dose of 50 mg/kgincreased the spontaneous locomotor activity (P〈0.05).At the dose of 10 mg/kg,it enhanced the hypnosis of mice induced by pentobarbital sodium (P〈0.05).In addition,it showed no impact on the coordinated locomotor ability.Conclusion Koumine inhibits mice's central nervous system at higher dose of its therapeutic range but may induce stimulatory effect beyond the therapeutic range after certain period of time.
