摘    要:目的:用新的修饰型吗呆代反义寡核苷酸(M-ODN)抑制肿瘤细胞bcl-2的表达,探讨发展新的抗肿瘤药物的可能性。方法:用吗啉代bcl-2反义寡核苷酸作用于K-562细胞,并与全硫代修饰的bcl-2反义寡核酸(S-ODN)为对比,MTT法观察细胞生长抑制率,流式细胞术观察细胞凋亡率。结果:吗啉代修饰型的bcl-2反义寡核苷酸在作用效率、持久性及低细胞毒性等方面均优于硫代修饰。结论:反义吗啉代寡核有望成为新的肿瘤基因治疗手段。

Abstract:Objective To investigate the anti-tumor effects of bcl-2 antisense morpholino oligodexynucleotide(M-ODN). Methods MTT spetrophotometry and flow cytometry were used to examine the effect of M-ODN on survival of K-562 cells and results were compared with that of phosphorothioate oligodexynucleotide(S-ODN). Results M-ODN offered many advantages over S-ODN in inhibitive efficacy,persistence and safety profile. Conclusion bcl-2 antisense M-ODN may become a new anti-tumor approach.
