急性髓性白血病体外药敏、p170表达和细胞代谢的关系

摘    要:目的了解急性髓性白血病(AML)体外药敏实验、p170表达和细胞代谢的关系.方法用MTT法检测27例AML患者体外药敏实验,同时检测其白血病细胞代谢和P170的表达,观察比较药敏结果、P170表达、细胞代谢之间及与临床化疗结果间的关系.结果体外药敏实验与临床治疗效果有相关性,二者的总符合率81.9%,阳性符合率88.9%,阴性符合率66.7%.细胞代谢与临床化疗疗效相关,细胞代谢高,对化疗反应好,缓解率高;细胞代谢与体外药敏实验和P170表达无相关性.P170表达与临床疗效相关,P170表达阳性组缓解率低于表达阴性组.结论MTT体外药敏实验、P170表达和细胞代谢可以辅助提高临床判断耐药的准确性,可为临床选药提供依据.

Abstract:Objective:To evaluate the relationship of chemosensitivity assay, blast cell metabolism and expression of p170 in acute myeloid leukemia patients.Methods:We examined the chemosensitivity and blast cell metabolism with MTT method and the expression of p170 with immunocytochemistry simultaneously in 27 cases, compared the correlation between outcome of chemotherapy and chemosensitivity assay, blast cell metabolism activity and expression of p 170.Results:The chemosensitivity assay correlated well with the outcome of the treatment with an overall predicated rate of 81.9%, appositive predictive rate of 88.9%, and a negative rate 66.7% respectively. There is a positive correlation between blast cell metabolism and remission induction. Patients with metabolism active cells had a better chance of achieving complete remission. No results showed that blast cell metabolism activity correlated with the chemosensitivity assay and expression of p170.Patients with positive expression of p170 showed a significant lower rate of complete remission than those with negative expression of p 170.Conclusions:Chemosensitivity assay, blast cell metabolism and expression of p170 are useful parameters to predict the outcome of the treatment.
