摘    要:目的验证不同浓度的辛伐他汀对人类成骨细胞(OB)功能表达和体外成骨能力的影响,研究雌激素促进成骨作用的机制及剂量效应关系.方法取成人的髂骨松质骨,采用胶原-胰蛋白酶消化法,获得松质骨中的成骨细胞,进行纯化和培养.在此基础上添加不同浓度的辛伐他汀(1×10-10,1×10-9,1×10-8,1×10-7,1×10-6,1×5-5mol/L)并进行培养.用生化法测定细胞碱性磷酸酶(ALP)活性,用放射免疫法测定细胞培养液中骨钙素(OC)和细胞间质的钙含量.结果辛伐他汀对成骨细胞ALP活性和骨钙素分泌的影响均呈正相关,即各浓度辛伐他汀对ALP活性、骨钙素的分泌和成骨细胞的成骨能力均有刺激作用,并与辛伐他汀剂量呈正相关.结论辛伐他汀能增加成骨细胞的ALP活性和骨钙素的产生,提高成骨细胞的成骨能力.

Abstract:Objective: To verify the effects of different concentration of simvastain on function of osteoblast bone formation and power of bone-grown up in vitro. Methods: Iliac spongy bone specimens were obtained from twelve adult patients scheduled surgery. After the bone pieces were treated with collagenasetrypsin, osteoblasts were obtained from cancellous bone. Then the osteoblasts were purified and cultivated. 0steoblasts were cultured in medium with simvastain of various concentrations (1×10-10, 1×10-9, 1×10-8, 1×10-7,1×10-6 and 1×5-5 mol/L). ALP activity, osterocalcin and power of bone-grow up were examined by biochemical assay and radioimmunologic assay (RIA) respectively. Results: Results showed that all above concentration of simvastain on activity and osteoblasts were positive. Simvastain of lower concentrations (1×10-10, 1×10-9, 1×10-8mol/L) showed the stimulated effect on the power of bone-grown up and that of higher concentration was not. Conclusions: Statins can increase osteoblast ALP activity, osteocalcin production and power of bonegrown up.
