透骨草提取物对人肝癌SMMC-7721细胞p53和PUMA蛋白表达的影响

摘    要:目的观察透骨草提取物对人肝癌SMMC-7721细胞形态和p53和p53上调凋亡调控因子(PUMA)蛋白表达的影响,探讨透骨草提取物诱导SMMC-7721细胞凋亡的分子机制。方法用倒置显微镜观察透骨草提取物对SMMC-7721细胞形态的影响;采用免疫组化方法检测透骨草提取物对SMMC-7721细胞p53和PUMA蛋白表达的影响。结果倒置显微镜观察可见,40.91μg/mL透骨草提取物处理的SMMC-7721细胞体积减小,形态轮廓不清晰,细胞皱缩变形,细胞脱壁增加。免疫组化检测结果显示,40.91μg/mL透骨草提取物处理的SMMC-7721细胞p53蛋白和PUMA蛋白表达均明显高于与对照组(P<0.01)。结论透骨草提取物可能通过上调p53和PUMA蛋白表达,诱导SMMC-7721细胞凋亡。

Abstract::Objectlve To observe the effects of Phryma leptostachya L. vat. asiatica Hara extract on the morphology and the expressions of p53 and PUMA proteins in human hepatocellular carcinoma SMMC-7721 cells,in order to investigate the molecular mechanism for Phryma leptostachya L. var. asiatica Hara extract inducing SMMC-7721 cell apoptosis. Methods The morphology of SMMC-7721 cells was observed with the inverted microscope. Immunohistochemistry technique was used to determine the expressions of p53 and PUMA proteins of SMMC-7721 cells. Results After SMMC- 7721 cells were treated with 40. 91 μg/mL Phryma leptostachya L. var. asiatica Hara extract, the changes of the volume of SMMC-7721 cell reduced, morphological profile became unclear, cell shrinkage was de- formed, and the number of exfoliative cells increased were observed with inverted microscope. Immunohistochemistry technique showed that the expressions of p53 and PUMA proteins of SMMC-7721 cells treated with 40.91 μg/mL Phryma Leptostachya L. var. asiatica Hara Extract increased than control group (P 〈 0. 01). Conclusion Phryma Leptostachya L. var. asiatica Hara Extract could induce the apoptosis of SMMC- 7721 cells by upregulating the expressions of p53 and PUMA proteins.
