高效液相色谱-直流安培法测定扶本增脉颗粒中丹酚酸B的含量

摘    要:目的:采用高效液相色谱-直流安培检测法建立扶本增脉颗粒中丹酚酸B的含量测定方法。方法:采用ODS-BP C18柱(200mm×4.6mm,5μm);以乙腈和0.1%磷酸(35:65,v/v)为流动相(流速1.0mL/min)进行等度洗脱;工作电极为Pt,参比电极为Ag/AgCl,工作电压为1.0V;柱温为30℃。结果:丹酚酸B质量浓度分别为0.005-10mg/L时与其峰面积呈良好的线性关系(r2=0.9998),回收率为97.8%-100.4%;相对标准偏差(RSD)5.0%,检测限为2μg/L。结论:该方法与高效液相色谱-紫外检测法相比,含量测定结果一致,检测限更低,是一种快速、灵敏、准确的分析方法,可以为扶本增脉颗粒的质量控制提供参考。

Abstract:Objective: To establish a rapid and accurate method for determination of salvianolic acid B in Fubenzengmai Granules by high performance liquid chromatography coupling with direct current amperometric detection.Methods: The separation was performed on an ODS-BP C18 column(200mm×4.6mm,5μm) by gradient elution with acetonitrile and 0.1% aqueous phosphoric acid as mobile phase(35:65,v/v) at a flow rate of 1.0mL /min.The work electrode,reference electrode and work potential were Pt,Ag/AgCl,1.0 V,respectively.The temperature of column was set at 30℃.Results: Calibration curve showed good linearity and the correlation coefficients(r2) was 0.9998.Average recovery of salvianolic acid B was 97.8-100.4% and relative standard deviation(RSD) was less then 5.0%,detection limit was 2μg/L.Conclusion: The result was as same as that determined by HPLC-UV,this method has lower detection limit,and appeared to be stable,sensitive and reproducible for determination of salvianolic acid B in Fuben Zengmai Granules.The study provided a scientific basis for the quality control of Fuben Zengmai Granules.
