摘    要:目的 :探讨胰岛素样生长因子 II(IGF II)对卵巢癌细胞SKOV3的基质金属蛋白酶 9(MMP 9)、金属蛋白酶组织抑制剂 1(TIMP 1)表达的影响。方法 :细胞 (1.5× 10 5/ml)培养 4 8h后 ,加入不同浓度的IGF II ,再培养 18h。用RT PCR法测定MMP 9mRNA、TIMP 1mRNA水平 ,用明胶酶谱法半定量测定MMP 9蛋白水平的变化。结果 :IGF II可刺激MMP 9蛋白的分泌。随着IGF II浓度增加 ,诱导作用增强 ;浓度较高时 ,MMP 9的分泌量轻度下降。但IGF II不影响MMP 9和TIMP 1的基因表达水平。结论 :IGF II可刺激卵巢癌细胞SKOV3中MMP 9的分泌 ,在卵巢癌的侵袭转移方面可能起重要作用

Abstract:Objective:To investigate the effect of IGF II on MMP 9 and TIMP 1 expressions in ovarian cancer cell line SKOV3.Methods:SKOV3 cells (1.5×10 5/ml) were cultured for 48 hours and then treated with IGF II at different concentration for additional 18 hours culture.MMP 9 secretion in conditional medium was analyzed by gelatin zymography.MMP 9 and TIMP 1 mRNA were measured by RT PCR.Results:The secretion of MMP 9 in SKOV3 increased after the treatment of IGF II.This stimulation was enhanced with increasing concentration of IGF II.However,treated with higher IGF II concentration (100ng/ml),MMP 9 secretion slightly decreased.Nevertheless,the expressions of MMP 9 and TIMP 1 mRNA had no significant response to IGF II treatment.Conclusion:IGF II may play an important role in the process of invasion and metastasis in ovarian cancer through increasing MMP 9 secretion.
