FK506抑制兔同种异体角膜缘移植免疫排斥反应的实验研究

目的 探讨FK506滴眼液对同种异体角膜缘移植术后排斥反应的免疫抑制作用。方法 新西兰兔48只48眼建立角膜缘缺陷症动物模型。随机分为FK506组、环孢霉素A组及生理盐水对照组3组，每组16眼。1个月后实施同种异体角膜缘移植术，眼表观察10周。结果 术后10周，对照组、环孢霉素A组、FK506组角膜新生血管指数平均秩分别为15．50、8．00、5．00。对照组角膜新生血管指数较其他2组高（P＝0．00）；而FK506组与环孢霉素A组间无显著差异（P＝0．12）；对照组上皮排斥反应发生最早，环孢霉素A组次之，FK506组最晚；术后10周，对照组、环孢霉素A组、FK506组移植排斥反应指数平均秩分别为13．67、8．07、4．58，对照组排斥反应指数较FK506组（P＝0．02）和环孢霉素A组（P＝0．00）高；环孢霉素A组移植排斥反应指数显著高于FK506组（P＝0．04）。同种异体角膜缘移植术后眼局部应用FK506滴眼液可延迟排斥反应发生，其免疫抑制效果优于环孢霉素A。

Experimental studies on the effects of the immunosuppressant FK506 on limbal allograft transplantation rejection on rabbits

Objective To discuss the immunosuppression of FK506 eye drops on limbal allograft transplantation rejection.Methods Limbal deficiency models were established in 48 New Zealand rabbits eyes,and were randomized into 3 groups, FK506 group ,CsA group and physiological saline control group,each having 16 eyes.Limbal allograft transplantations were adopted one month later.The ocular surfaces were observed clinically for 10 weeks.Results Ten weeks after operation,corneal neovascularization index in the control group,CsA group and FK506 group were 15.50,8.00 and 5.00 respectively. It was no difference between FK506 group and CsA group( P =0.12), and it was higher in the control group than in the other two groups( P =0.00).Epithelial rejection occurred earliest in the control group,occurred earlier in CsA group than in FK506 group.Rejection index in the control group,CsA group and FK506 group were 13.67,8.07 and 4.58 respectively.It was higher in the control group than that in FK506 group( P =0 02) and CsA group( P =0.00), and that in CsA group it was higher than that in FK506 group( P =0 04).Conclusion Topically administed FK506 eye drops early after limbal allograft transplantation can delay rejection response,its immunosuppression effects were better than CsA.
