丙氨瑞林对子宫内膜癌裸鼠移植瘤的生长及FHIT表达的影响

摘    要:目的观察醋酸丙氨瑞林对子宫内膜癌裸鼠移植瘤生长的抑制作用及可能的作用机制。方法将HEC-1-B细胞注入裸鼠左前肢近腋窝处皮下,选24只雌性荷瘤裸鼠随机分为4组,即低剂量组(18肛g/kg)、中剂量组(36μg/kg)、高剂量组(72μg/kg)和空白对照组(0μg/kg),采取肌肉注射方式给药4周。将瘤体完整取出,测量体积、绘制生长曲线、计算抑瘤率,并用免疫组织化学法检测瘤组织中FHIT蛋白的表达。结果①随着醋酸丙氨瑞林浓度由低到高,癌细胞受抑制渐趋明显,低、中、高剂量组抑瘤率分别为17.0%、24.1%、42.1%,且与对照组比较差异有统计学意义(P〈0.05)。②免疫组织化学结果显示,各干预组的FHIT蛋白表达水平较对照组明显升高,且组间差异有统计学意义(P〈0.05)。结论醋酸丙氨瑞林对子宫内膜癌皮下移植瘤生长有抑制作用,且呈剂量依赖关系.其机制可能与上调FHIT蛋白表达有关。

Effects of alarelin on subcutaneous xenograft of human endometrial carcinoma and its expression of FHIT in nude mice

Abstract:Objective To study if alarelin can inhibit nude mice bearing human endometrial carcinoma and to explore its possible molecular mechanisms.Methods Human endometrial carcinoma HEC-1-B cells were injected into the left axillary fossa of the nude mice,then the mice were randomly divided into four groups: low-dose group(18 μg/kg),middle-dose group(36 μg/kg),high-dose group(72 μg/kg) and blank control group(0 μg/kg).They were injected into the nude mice in the same way.After observing the systemic condition of the mice for 4 weeks,we measured the size,drawed the growth curve,calculated the inhibition rate of the tumor,then detected the expression of FHIT with the immunohistochemical technology.Results ①The inhibition rate of the tumor increased obviously with the rising of alarelin concentrations.18,36,72 μg/kg of alarelin inhibted tumor by 17.0%,24.1%,42.1%.Compared with the control group,the differences were significant(P < 0.05,respectively).②The immunohistochemical results showed that the expression of FHIT rised obviously in treated groups with the increasing dose of alarelin.Compared with the saline group,there were statistically significant differences(P < 0.05).Conclusion Alarelin might inhibit directly the growth of subcutaneous xenograft of human endometrial carcinoma in nude mice and the effect is dose-dependent.And it may be related to the up-regulation of FHIT protein.
